Engineered cross‐reacting nanobodies simplify comparative oncology between humans and dogs

Abstract: Antibodies cross-reacting with homologue antigens in different species would be essential reagents for the development of comparative oncology studies. In comparison with conventional immunoglobulin Gs, recombinant nanobodies (single-domain variable regions of heavy-chain only antibodies of Camelidae origin) can be easily isolated in vitro and engineered into a variety of reagents with optimized characteristics for different research and clinical applications. We recovered an anti-human epidermal growth factor… Show more

“…Recognition of HER2 and DER2 was evaluated by flow cytometry and IFA to detect breast cancer cells from humans (SKBR3) and dogs (SH1B and P114). Therefore, these rAbs can be used to identify malignant cells and have the potential to be used as immunotherapeutics in dogs and humans ( 28 ).…”

“… 40 Therefore, we decided to explore the short H6 tag, fused to nanobodies and GFP ( Figure 1 B), as a tool to generate large self-assembled multivalent and multifunctional nanoparticles. For that, two unrelated nanobodies were selected as models, namely A3C8, specific for ricin toxin, and EM1, specific for Her2 ( Figure 1 C), 41 , 42 and engineered to demonstrate the feasibility of the proposed approach, that is, controlled oligomerization into nanoparticles while preserving functionalities of the nanobodies and of other incorporated protein domains.…”

“…In terms of tumour treatment, advantage was taken of high similarity between human epidermal growth factor receptor and its canine homologue and two recombinant antibodies with crossreactivity generated; both seemed suitable for detection of malignant cells when tested to detect breast cancer cells (132).…”
