Endowing iPSC-Derived MSCs with Angiogenic and Keratinogenic Differentiation Potential: A Promising Cell Source for Skin Tissue Engineering

Abstract: Induced pluripotent stem cells (iPSC) hold tremendous potential for personalized cell-based therapy for skin regeneration. Aiming to establish human iPSCs as a potential cell source for skin tissue engineering, we expect to obtain an epidermal-like cell line with angiogenic and keratinogenic differentiation potential via inducing iPSC-derived mesenchymal stem cells (iPSC-MSCs) with basic fibroblast growth factor (bFGF) and/or keratinocyte growth factor (KGF). The results show that iPSC-MSCs were successfully i… Show more

“…We speculated that this is because the concentration of released growth factors increases as the concentration of PRP and LyPRP increased. With prolonged culturing time, the cells tended to proliferate rather than differentiate, so the expression of osteogenic differentiation related genes is slightly reduced (W. Lin et al, ; Shirai, Ishisaki, Kaku, Tamura, & Furuichi, ).…”

“…iPSCs are a new class of pluripotent stem cells which can differentiate into cells of all three germ layers 58 . These cells are generated by reprogramming somatic cells (e.g., dermal fibroblasts) via exposure to four specific genes encoding transcription factors, that is, octamer 3/4 (Oct3), sex‐determining region Y‐box (Sox2), Kruppel‐like factor 4 (Klf4), and v‐Myc myelocytomatosis viral oncogene homologue (c‐Myc) 59 .…”

“…Cell‐based therapy offers a promising strategy for tissue regeneration (Lin et al, ; Mount, Ward, Kefalas, & Hyllner, ). Compared with cells alone, 3D biomaterials can serve as ideal scaffolds for cell delivery, protecting cells from injection shear, providing structural support, and controlling ideal cell fate, thereby improving the outcome of cell‐based therapy (Khademhosseini & Langer, ).…”
