Endothelial Cell-Secreted EGF Induces Epithelial to Mesenchymal Transition and Endows Head and Neck Cancer Cells with Stem-like Phenotype

Abstract: Emerging evidence suggests that endothelial cell-secreted factors contribute to the pathobiology of squamous cell carcinoma (SCC) by enhancing invasive migration and resistance to anoikis. Here we report that SCC cells within the perivascular niche have undergone epithelial to mesenchymal transition (EMT) in a primary human SCC of a patient that developed distant metastases. Endothelial cell-secreted EGF induced EMT of human SCC cells in vitro and also induced acquisition of a stem-like phenotype. In vivo, tum… Show more

“…The value of CD44 as a marker of OSCC progression and prognosis is also controversial. Certain studies in HNSCC have reported aldehyde dehydrogenase (ALDH) + cells with typical CSC behavior, in particular the tumorigenic ability (55)(56)(57)(58). The specificity of CD44 as a marker of CSCs in HNSCC is reported to be increased in combination with ALDH (55,56).…”

“…There are also changes in cellular proteins, such as integrins and a-smooth muscle actin, as well as ECM proteins, such as collagen, laminin, and fibronectin. 149,[213][214][215][216][220][221][222][223] The HNSCC cells that undergo EMT have been shown to be more invasive. * The loss of E-cadherin associated with EMT is an indicator of poor prognosis in HNSCC.…”

“…225,226 Cancer stem cells exhibit the properties of self-renewal, potential to behave as tumor progenitor cells, and the ability to differentiate. 223,[225][226][227] Cancer stem cells are slow-dividing and therefore often more resistant to chemotherapy. [225][226][227] Cancer stem cells are found in invasive fronts, and in many tumors are near the blood vessels.…”
