Mechanisms of Invasion in Head and Neck Cancer

Abstract: Context.-The highly invasive properties demonstrated by head and neck squamous cell carcinoma (HNSCC) are often associated with locoregional recurrence and lymph node metastasis in patients and is a key factor leading to an expected 5-year survival rate of approximately 50% for patients with advanced disease. It is important to understand the features and mediators of HNSCC invasion so that new treatment approaches can be developed.Objectives.-To provide an overview of the characteristics, mediators, and mecha… Show more

“…ERK2 (encoded by MAPK1) is an important protein in the MAPK signaling pathway regulating cell proliferation, differentiation, and migration. Aberrant and/or persistent activation of the MAPK cascades can lead to the development and invasion of tumors including HNSCC [41][42] . The positive association between ERK2 expression and tumor maximum 3D diameter indicates larger tumors tend to have a higher ERK2 expression.…”

“…Oral cancer is one of the top 10 prevailing cancers worldwide, showing high morbidity and mortality 1 . Poor prognosis in oral cancer is mainly attributed to its tendency for highly local invasion 2 and recurrence rate 3 . Metastasis consists of several processes, including cancer cell proliferation, angiogenesis, migration, invasion, and microenvironment interaction 4 .…”

“…More than half of sporadic HNSCCs are diagnosed at locally advanced or metastatic stages, and approximately 50% of treated patients relapse with local or distant metastasis, both bearing poor rates of remission (1, 2). Unfortunately, decades of research have not improved HNSCC outcomes significantly, and the classic therapeutic option, a combination of surgery, radiation and chemotherapy, leaves patients permanently disfigured.…”
