“…The founding member of the family, Commd1, was originally identified as the causative gene in terrier dogs with copper toxicosis, and interacts with and traffics the Wilson Disease copper transport protein ATP7B and its homologue ATP7A . The assembly of the CCC sub‐complex is facilitated by the binding of the COMM domains of the COMMD proteins to the N‐terminal NN‐CH domains of CCDC22 and CCDC93 . The CCDC22‐CCDC93 complex also interacts with Retriever, as well as the WASH complex subunit FAM21 which is required for recruitment of the Commander complex to endosomes .…”

“…The propensity to assembly into higher order oligomers is a noted feature of the COMMD proteins. For example, all COMMD proteins were identified in a > 600 kDa complex with other Commander subunits after cell lysis and fractionation, heterotypic interactions between Commd1, Commd6, Commd5 and Commd9 were identified using purified proteins, and a specific analysis of a purified Commd5‐Commd10 heterodimer revealed the existence of a tetrameric 2:2 stoichiometry . Furthermore, many of the proteins show inter‐dependence in their stability as knockouts of individual COMMD or CCDC genes typically leads to loss of other CCC complex subunits .…”

“…It is of interest that WASH also regulates the recycling of SNX17 cargoes including the integrin α5β1, EGFR, LDLR and Notch ligand Jag1. However, how SNX17 couples trafficking of its cargos to WASH‐regulated subdomains remained a mystery until very recently.…”

“…COMMD1 has been the focus of most studies to date and much less is known about the function of other members of the family. All models of Commd gene deletion studied thus far in mice result in embryonic lethality (Li et al, 2015;. Detailed characterization of the embryonic phenotypes revealed non-overlapping developmental defects and highlighted unique functions for these factors including a role for COMMD1 in HIF regulation (van de Sluis et al, 2010;, and the identification that COMMD9 regulates developmental events through regulation of Notch trafficking (Li et al, 2015).…”
