Enantioseparation of 1-phenyl-1-propanol by supercritical fluid-simulated moving bed chromatography

The effect of pressure drop on the performance of supercritical fluid chromatography systems using a modified mobile phase (carbon dioxide + ethanol) was studied. Experiments were performed on a Lichrospher-RP-18 column with phenanthrene as a solute. A wide range of back pressures (130 to 210 bar) and modifier concentrations (2 to 7% w/w) have been explored. Experiments yielding both small and large pressure drops were performed. From these experiments, parameters to describe pressure drop, retention, and column efficiency were extracted, and were used to simulate the dynamics of the chromatographic column. A good match between the experimentally measured and calculated values of pressure drop, retention times, and column efficiency was observed. At low back pressure and modifier composition, significant loss of column efficiency was observed.

Die Verfügbarkeit reiner Enantiomere ist von steigender Bedeutung für die pharmazeutische Industrie sowie auch für die Agrochemie und Biotechnologie. Generell gibt es zwei konkurrierende Ansätze zur Gewinnung von reinen Enantiomeren. Der “chirale” Ansatz basiert auf der Entwicklung einer asymmetrischen Synthese, die selektiv eines der Enantiomere liefert. Der “racemische” Ansatz basiert auf der Trennung eines Gemischs der beiden Enantiomere, wobei hier in den letzten Jahren bemerkenswerte Fortschritte erzielt werden konnten. Dieser Aufsatz fokussiert insbesondere auf enantioselektive Kristallisationsprozesse und präparative Chromatographie, einschließlich hybrider Trennprozesse und der zusätzlichen Integration von Racemisierungsschritten. Zur Illustration dienen mehrere in unserem Arbeitskreis untersuchte Trennungen.

New Developments in Simulated Moving Bed Chromatography

Chromatographic separation processes are widely used to isolate and purify value added products. Most frequently, such separation processes are performed exploiting the principles of injecting samples of the feed mixture in a repetitive periodic manner and collecting the target products batchwise. In the early 1960s, an alternative operation principle based on using several columns connected in series and exploiting a continuous countercurrent movement between the mobile and stationary phases initiated significant improvements in performing specific separation processes in the petrochemical and sugar industries. In the last decade, the so-called simulated moving bed (SMB) chromatography has rapidly entered the pharmaceutical industry. In particular enantioseparations using chiral stationary phases have become a strong driving force to develop more sophisticated concepts of continuous chromatography. Currently, the growing interest in efficient methods for the downstream processing of biomolecules enhances interest in SMB processes.
