Employing an R Software Package rsm for Optimizing of Genistein, Daidzein, and Glycitein Separation and Its Application for Soy Milk Analysis by HPLC Method

Abstract: Soy milk, one of the soybean products, become more popular in recent years due to its benefit for human health. Biological activities of soybean products have been widely studied according to the presence of isoflavone aglycones content, including genistein, daidzein, and glycitein. Hence, it is important to develop an effective and efficient analytical method to provide guidance regarding appropriate isoflavone intake levels for soy milk. A reversed-phase high performance liquid chromatography (HPLC) method w… Show more

“…Box-Behnken Design (BBD) is one of the optimization methods recently used in the nanoemulsion formulation area [24][25][26][27]. BBD allows the formulators to optimize more than two factors of formulation in three different levels to obtain multiple response optimization [28][29]. BBD has a distinct advantage compared to the central composite design (CCD) in a way that BBD requires fewer number of points; therefore, it is more economical and more efficient [30].…”

“…The method referred to an HPLC method developed by Dwiastuti et al [31] in analyzing 4-n-butyl resorcinol in the nanosystem formulation. An open-source R software package called "rsm" was also applied to economically implement a BBD model for computer-aided optimization purposes [29]. [32].…”

“…HPLC optimization followed by quantitative estimation was previously reported using single-point calibration [32,47]. In this study, a multiple-point calibration method was applied for estimating the content of resorcinol and 4-n-butyl resorcinol.…”

“…Optimization of separation conditions is a crucial stage in analytical method development [32][33][34]. Chemometrics-assisted chromatographic optimization can be performed using response surface methodologies (RSM) such as factorial, central composite (CCD), Box-Behnken (BBD), and Doehlert designs [35].…”

“…Response surface methodology (RSM) is based on combining mathematical and statistical techniques to set a polynomial equation for experimental findings. It can explain the performance of a response affected by several variables, evaluate the most effective factor on the selected response, create a model for prediction, and explore experimental conditions to the optimization (Riswanto et al, 2019;Chollom et al, 2020). Hence, RSM has been utilized in several recent agricultural, pharmacological, and chemical studies and related fields (Abdella et al, 2018;Kaur et al, 2018;Yoo et al, 2018;Ning and Yue, 2019).…”
