Qa-2, a murine class Ib major histocompatibility complex (MHC) molecule, is a possible functional homolog of human leukocyte antigen G (HLA-G). Both molecules have been implicated in immunoregulation and embryonic development and both occur in membrane-bound and soluble isoforms that arise by alternative splicing. Soluble splice variants have been implicated in the reproductive functions of HLA-G. While soluble variants of Qa-2 have been previously detected in T lymphocytes, we now demonstrate the presence of mRNA for one of the two known soluble forms of Qa-2 in eight-cell embryos and in blastocysts. Qa-2 is glycosylphosphatidy-linositol (GPI) linked in the outer leaflet of the cell membrane and is found in lipid raft microdomains where other raftassociated proteins transduce signals into the cell. In contrast, HLA-G has a truncated six amino acid cytoplasmic tail. By fluorescence co-localization in JEG-3 cells, using fluorescent cholera toxin β subunit (a lipid raft marker) and anti-HLA-G antibody, we have demonstrated that membrane-bound HLA-G also localizes to lipid rafts, consistent with functional homology between the two molecules. Finally, our experiments in which we have purified Qa-2 and transferred it via a process known as protein painting to Qa-2 negative cells represent a model for potential therapy involving HLA-G.

Purpose : To find an effective method to select embryos from ICSI with better chromosomal status when preimplantation-genetic-diagnosis (PGD) is not applied. Methods : Several morpholological evaluations were done in same embryos. Embryos from ICSI were classified on day 1 according to pronuclear-nucleoli arrangement. On day 3, classification was done according to number, fragmentation, size and shape of cells. In some patients, embryos exhibiting good quality on day 3 (at least six regular blastomeres with cell fragmentation lower than 20%) were also submitted to PGD, irrespective to pronuclearnucleoli morphology. Results : Forty-two per cent of normally fertilized embryos showed pronuclear-nucleoli-goodmorphology; from those, 86% were also classified as good quality on day 3. Good-quality embryos submitted to PGD have shown lower chromosomal abnormality rates when also classified as pronuclear-nucleoli-good-morphology. Conclusions : Pronuclear-nucleoli morphology seems to be correlated with PGD results. This criterion may prove useful for pre-select embryos with normal chromosomal package when PGD is not applied.

The risk to maternal-foetal health due to multiple pregnancies can be decreased by reducing the number of transferred embryos. Therefore, new tools for selecting embryos with the highest implantation potential should be developed. The aim of this study is to evaluate the predictive implantation ability of morphological and morphometric variables by analyzing images of embryos. This is a retrospective study of 135 embryo photographs from 112 IVF-ICSI cycles performed between January and March 2011. The embryos were photographed immediately before transfer using "Cronus 3" software and their images were analyzed using the public program ImageJ.Significant effects and higher discriminant power to predict implantation were observed for the morphometric embryo variables in comparison with morphological ones. The features for successfully implanted embryos were: 4-cells on day 2 of development; all blastomeres with circular shape (roundness factor greater than 0.9) ), an average ZP thickness of 13 microns and an average of 17695.1 microns 2 for the embryo area. The size of the embryo, which is described by its area and the average roundness factor for each cell, provides two objective variables to consider when predicting implantation. This method would improve the current "embryo classification systems".
