ELISA for the detection of the triazole fungicide myclobutanil.

Abstract: A competitive enzyme-linked immunosorbent assay (ELISA) system for quantitative detection of myclobutanil was developed. Polyclonal antibodies were raised in rabbits against the haptenic compound 2-(4-chlorophenyl)-2-[(1,2,4-triazol-l-yl~methyll hexanoic acid, conjugated to conalbumin through the carboxylic acid moiety. The standard curve based on a homologous system using the above hapten conjugated to bovine serum albumin displayed a detection range from 0.3 to 200 pgl mL for myclobutanil with an IC50 value … Show more

“…Myclobutanil (MT), (RS)-2-(4-chlorophenyl)-2-(1,2,4-triazol-1-ylmethyl) hexanenitrile, is a typical triazole fungicide used for the control of powdery mildew and scabbing of plants. 5 Fungicidal action occurs through inhibition of the sterol 14-demethylase enzyme. 6 It has a single chiral center and consists of two enantiomers.…”

“…Several antibody-based tests for triazole fungicides have been developed, including for triadimefon, tetraconazole, penconazole, myclobutanil, tebuconazole and hexaconazole (Chen, Dwyre-Gygax, Hadfield, Willetts, & Breuil, 1996;Danks, Chaudhry, Parker, Barker, & Banks, 2001;Forlani, Arnoldi, & Pagani, 1992;Newsome, 1986;Szekacs & Hammock, 1995). To date, no immunoassay for diniconazole has been reported.…”

“…The pH value may play a significant role in molecular adsorption onto metallic surfaces. 27 The p K a (logarithm of the dissociation constant) values of triazole 28 and MCB 29 are 2.2 and 2.3, respectively. We found that MCB exhibited strong SERS spectra in the Ag NPs only under acidic conditions at pH values below 5 (see Fig.…”
