Electrophoretic Assembly of Antibody–Antigen Complexes Facilitates 1000 Times Improvement in the Limit of Detection of Serological Paper-Based Assay

Abstract: Serological assays detect the presence of specific antibodies in blood. There are urgent and important applications for serological point-of-care (POC) assays. However, available detection methods are either insufficiently sensitive or too complex for POC settings. Here, we demonstrate that lateral flow immunoassay (LFIA), which is arguably the simplest universal molecular detection approach, can serve as a methodological platform for highly sensitive serological POC assays if combined with a simple, fast, and… Show more

“…This blocking mechanism was reached via electrokinetically to drive delivery of the label once the antigen-antibody is in the test zone. The limit of detection using electrophoretically driven LFIA (eLFIA), has been proven to be 1000 times lower than that of conventionally LFIA [55]. Various post-assay upgrades based on chemical reactions to facilitate high sensitivity for LFIA have been presented in a critical review [56].…”

“…15 showcases several examples of colorimetric, electrochemical, and fluorescence detection methods. The combination of colorimetric and electrochemical techniques 170 has demonstrated a 10 3 -fold lower LoD than ordinary lateral flow assays, resulting in successful clinical serological implementation. This improvement in the LoD can be attributed to the electrostacking of gold nanoparticle-labeled proteins.…”
