Ehrlichia SLiM ligand mimetic activates Hedgehog signaling to engage a BCL-2 anti-apoptotic cellular program

Abstract: Ehrlichia chaffeensis (E. chaffeensis) has evolved eukaryotic ligand mimicry to repurpose multiple cellular signaling pathways for immune evasion. In this investigation, we demonstrate that TRP120 has a novel repetitive short liner motif (SLiM) that activates the evolutionarily conserved Hedgehog (Hh) signaling pathway to inhibit apoptosis. In silico analysis revealed that TRP120 has sequence and functional similarity with Hh ligands and a candidate Hh ligand SLiM was identified. siRNA knockdown of Hh signalin… Show more

“…Infectious dense-cored E. chaffeensis with T1SS TRP120 effector on the surface binds (1) to the ligand binding region (LBR) of the Notch-1 receptor via the Notch ligand memetic motif (Patterson, Velayutham et al 2022) and activates Notch signaling. (2) NICD translocates to the nucleus and activates Notch gene transcription and (3) binds directly to XIAP prevent XIAP degradation, (4) inhibiting Caspase-3, −7 and −9 activation. (5) Active Caspase-3 may also cleave XIAP into two fragments: BIR1 and −2 and BIR3-RING.…”
