Eficiência in vitro de acaricidas sobre carrapatos de bovinos no Estado do Rio Grande do Sul, Brasil

This study was conducted to obtain an epidemiological view of acaricide resistance in populations of Rhipicephalus (Boophilus) microplus in the State of Mato Grosso do Sul. Twenty-four tick samples were collected from municipalities in the State where farmers had reported concerns about resistance to or failure of tick control. These ticks were subjected to in vitro resistance detection assays using the adult immersion test (AIT). The efficacy of alpha-cypermethrin, cypermethrin and amitraz treatments on samples collected throughout the State was generally poor. AIT showed efficacy ≥ 90% from the use of DDVP + chlorfenvinphos) (20 out of 21 municipalities), dichlorvos + cypermethrin (10 out of 16 municipalities) and cypermethrin + citronella + chlorpyrifos + piperonyl butoxide (20 out of 21 municipalities). PCR assays were used to detect the presence of pyrethroid resistance-associated sodium channel gene mutation. Larvae from three different populations that had previously been diagnosed as pyrethroid-resistant, through AIT, were evaluated. The PCR assays showed that the pyrethroid resistance-associated gene mutation was absent from these three populations. This study confirms that the emergence of resistance is a constant challenge for the livestock industry, and that development of resistance continues to be a major driver for new antiparasitic drugs to be developed.

The synthetic pyrethroids and their associations have been widely used for controlling Rhipicephalus (Boophilus) microplus. The frequent use of acaricides has been inducing the development of resistance in the tick populations. The aim of this study was to assess the susceptibility of R. (B.) microplus populations to pyrethroids and their associations in the region of Garanhuns, Pernambuco, Brazil. In addition, the level of information among farm owners regarding tick control measures was investigated. Ticks were collected directly from naturally infested dairy cattle in the region and were exposed to pyrethroids and their associations. At the same time, an epidemiological questionnaire was applied with the aim of investigating the level of information among the farmers. The results reported here indicate that R. (B.) microplus populations in the dairy region of Garanhuns show resistance to pyrethroids and their associations, except when the product is associated with piperonyl butoxide. Regarding the results from the epidemiological survey, it was seen that there is a considerable lack of information among the farmers in relation to ixodid control measures. The level of ticks resistance to acaricides varied widely across the region studied. No alternative control programs have been implemented among these farms, thus demonstrating that there is a need for more information relating to the biology and control of R. ResumoOs piretróides sintéticos e associações são acaricidas amplamente utilizados para o controle do Rhipicephalus (Boophilus) microplus. O uso frequente destes fármacos tem induzido o surgimento de populações de carrapatos resistentes. Objetivou-se neste estudo avaliar a susceptibilidade de populações de R. (B.) microplus aos piretróides e associações. Além disso, investigou-se o nível de informações dos proprietários com relação às medidas de controle utilizadas. Os carrapatos foram coletados diretamente de bovinos naturalmente infestados na região leiteira de Garanhuns, Pernambuco e expostos a piretróides e suas associações. Contemporaneamente, foi aplicado um questionário epidemiológico com o objetivo de avaliar o nível de informação dos produtores. Os resultados aqui obtidos demonstram que populações de R. (B.) microplus provenientes da região leiteira de Garanhuns apresentam resistência aos piretróides, exceto quando o produto está associado com butóxido de piperonila. Em se tratando dos resultados referentes ao inquérito epidemiológico, verificou-se considerável falta de informação por parte dos produtores com relação às medidas de controle de ixodídeos. Assim, verifica-se que o nível de resistência aos acaricidas é muito variável na região estudada e que não há, nas propriedades, a adoção de programas alternativos de controle, demonstrando a necessidade de maiores informações relacionadas à biologia e ao controle do R. Comparison of three larval bioassays to evaluate susceptibility of Rhipicephalus (Boophilus) microplus to amitraz

Field samples of Rhipicephalus (Boophilus) microplus from the state of Rio Grande do Sul, Brazil, were assessed using the following methods: larval packet test (LPT), larval immersion test (LIT) and syringe immersion test (SIT). The following parameters were determined for each population and for the Mozo susceptible reference strain: lethal concentration for 50% (LC 50 ) with its 95% confidence interval (95% CI), regression line slope and resistance ratio (RR). Using the LPT, only one population was susceptible to amitraz, presenting a RR of 1.9. Using the same technique, the other populations presented RRs of between 92.9 and 3445.8 and were considered resistant. The LC 50 of the Mozo strain calculated using the LPT, LIT and SIT was 2.9, 27.3, and 52.7 µg/mL, respectively. In general, a good fit to the probit statistical model was only achieved using the LPT. The results obtained in this study impair recommendations for using the LIT and SIT to diagnose amitraz resistance in R. (B.) microplus populations. ResumoAmostras de Rhipicephalus (Boophilus) microplus coletadas à campo no Rio Grande do Sul, Brasil, foram analisadas pelos seguintes métodos: teste do pacote de larvas (TPL), teste de imersão de larvas (TIL) e teste de imersão em seringas (TIS). Os seguintes parâmetros foram determinados para cada população e para a amostra referência suscetível Mozo: concentração letal para 50% (CL 50 ) e seu intervalo de confiança de 95% (IC95%), inclinação da reta de regressão e os fatores de resistência (FR). Pelo TPL, apenas uma população foi sensível ao amitraz, com FR de 1,9. Utilizando a mesma técnica, as outras amostras apresentaram FR entre 92,9 e 3445,8 sendo consideradas resistentes. As CL 50 da cepa Mozo calculadas por meio do TPL, TIL e TIS foram 2,9, 27,3 e 52,7 µg/mL, respectivamente. De forma geral, a adequação ao modelo estatístico de probitos só foi alcançada com o uso do TPL. Os resultados obtidos no presente estudo limitam a recomendação de uso do TIL e TIS para diagnóstico de resistência ao amitraz em populações de R. (B.) microplus.
