Efficient Approaches to in vitro Multiplication of Lilium candidum L. with Consistent and Safe Access throughout Year and Acclimatization of Plant under Hot-Summer Mediterranean (Csa Type) Climate

Abstract: Lilium candidum L. is one of the most fragrant plant species among Liliaceae. The indigenous and natural populations of L. candidum are significantly affected by fast increasing anthropological population pressure and increased carbon fuel pollution. There is a necessity for development of a micropropagation protocol to serve in effective manner for both commercial multiplication and protection of the plant. The study targeted to develop a strategy for effective in vitro plant propagation system using static l… Show more

“…In that context, the combination of auxins and cytokinin's at concentrations below 2 mg L −1 has been reported to improve regeneration efficiency and bulb formation [2,8,22]. However, factors such as explant type and sucrose concentrations [23,24] and the presence of reserve elements, endogenous growth regulators, and genetic characteristics of the plant material must be taken into account [16].…”

“…In this study, it was observed that a simple activity such as covering the pots with polyethylene sheeting and maintaining adequate watering frequencies allowed the seedlings to remain unaffected by the ex vitro conditions, resulting in the survival of all seedlings and growth of up to 13 cm in the first 45 days. In this regard, Daneshvar-Royandazagh [2] and Saetiew and Umamanit [34] concluded that keeping seedlings in a moist environment during their first days of growth is critical to prevent losses.…”

“…Bulb scales planted on MS medium containing 5.37 µM NAA and 4.44 µM N-6 benzylaminopurine (BAP) stimulated a 100% growth percentage with 4.3 shoots per explant and 1.4 microbulbs of Lilium longiflorum . A single scale of Lilium candidum planted on MS medium with 0.23 µM TDZ and 1.07 µM NAA resulted in 6.5 bublets per explant and 26 bublets per baby food culture jar were regenerated on the similar medium, rooted ½ MS medium fortified by 3.2 µM IBA and successfully transferred ex vitro with high survivability (Royandazagh, 2019). Culturing leaf explants of Lilium orientalis 'Starfigther' on MS medium added by 2.27 µM TDZ and 45.24 µM 2,4-D regenerated 5.7 shoots after 1 month and significantly proliferated with 8.3 shoots/explant, 5.4 leaves/shoot and 8.8 scales/explant (Youssef et al, 2019).…”

“…The general methods of the propagation of horticultural varieties of lily can be divided into seed propagation and vegetative propagation. As vegetative propagation methods, propagation by bulblets and bulbils, scaling, and tissue culturing have been widely studied (Kim and de Hertogh, 1997;Daneshvar Royandazagh, 2019). Among these methods, scaling and tissue culturing are effective for mass reproduction.…”
