Abstract:The methanol extract of the aerial parts of the medicinal plant Pedicularis sibthorpii Boiss., Scrophulariaceae, growing in the Azerbaijan province of Iran, was found to be active in the 2,2-diphenyl-1-picryl-hydrazyl (DPPH) and the antibacterial agar well diffusion assays, but no general toxicity was observed in the brine shrimp lethality assay. A combination of solid-phase extraction (SPE) and preparative reversed-phase high-performance liquid chromatography (prep-RP-HPLC) analyses of the methanolic extract afforded three phenylethanoids (verbascoside, martynoside and isomartynoside), an iridoid (aucubin), a flavonoid (luteolin 7-O-β-D-glucopyranoside) and mannitol, and the structures of these compounds were elucidated unambiguously by spectroscopic means. The distribution of the isolated compounds within the genus Pedicularis has also been discussed.

Luteolin, which is structurally classified under the flavonoids, is naturally distributed abundant among medicinal plants, vegetables and fruits. The wound healing effects of Luteolin ointments were evaluated for incision and excision wound models on non-diabetic and Streptozotocin induced diabetic rats by using macroscopical, biomechanical, biochemical and histopathological methods. Two different concentrations (0.5 % and 1 % (w/w)) of the Luteolin ointment were prepared with the mixture of glycol stearate:propylene glycol:liquid paraffin (3:6:1) and 0.5 g of the ointments were applied topically on wounds once daily for 7 and 14 days. During the experiments, wounds were visually observed, photographically documented and wound areas were measured. After 7 and 14 days treatment, animals were sacrificed under anesthesia and hydroxyproline measurement and biomechanical analysis were performed. Histopathology of the wound area was evaluated considering features of re-epithelialization, thickness of the granulation tissue, angiogenesis, presence of inflammation, number of mast cells. Outcomes of this study revealed that Luteolin ointments improved wound healing process of skin tissue both in non-diabetic and diabetic wounds. The best wound healing activity was observed in incision and excision wounds (97.6 %, 96.1 %, respectively) treated with 0.5 % (w/w) Luteolin ointment on day 14 according to macroscopic results.

Luteolin as a modulator of skin aging and inflammation

Luteolin belongs to the group of flavonoids and can be found in flowers, herbs, vegetables and spices. It plays an important role in defending plants, for example against UV radiation by partially absorbing UVA and UVB radiation. Thus, luteolin can also decrease adverse photobiological effects in the skin by acting as a first line of defense. Furthermore, anti-oxidative and anti-inflammatory activities of luteolin were described on keratinocytes and fibroblasts as well as on several immune cells (e.g., macrophages, mast cell, neutrophils, dendritic cells and T cells). Luteolin can suppress proinflammatory mediators (e.g., IL-1β, IL-6, IL-8, IL-17, IL-22, TNF-α and COX-2) and regulate various signaling pathway (e.g., the NF-κB, JAK-STAT as well as TLR signaling pathway). In this way, luteolin modulates many inflammatory processes of the skin. The present review summarizes the recent in vitro and in vivo research on luteolin in the field of skin aging and skin cancer, wound healing as well as inflammatory skin diseases, including psoriasis, contact dermatitis and atopic dermatitis.In conclusion, luteolin might be a promising molecule for the development of topic formulations and systemic agents against inflammatory skin diseases.
