Root rot disease is a serious infection leading to production loss of Chinese wolfberry (Lycium barbarum). This study tested the potential for two bacterial biological control agents, Bacillus amyloliquefaciens HSB1 and FZB42, against five fungal pathogens that frequently cause root rot in Chinese wolfberry. Both HSB1 and FZB42 were found to inhibit fungal mycelial growth, in vitro and in planta, as well as to promote the growth of wolfberry seedlings. In fact, a biocontrol experiment showed efficiency of 100% with at least one treatment involving each biocontrol strain against Fusarium oxysporum. Metagenomic sequencing was used to assess bacterial community shifts in the wolfberry rhizosphere upon introduction of each biocontrol strain. Results showed that HSB1 and FZB42 differentially altered the abundances of different taxa present and positively influenced various functions of inherent wolfberry rhizosphere bacteria. This study highlights the application of biocontrol method in the suppression of fungal pathogens that cause root rot disease in wolfberry, which is useful for agricultural extension agents and commercial growers.

Plant growth-promoting rhizobacteria (PGPR) are specialized bacterial communities inhabiting the root rhizosphere and the secretion of root exudates helps to, regulate the microbial dynamics and their interactions with the plants. These bacteria viz., Agrobacterium, Arthobacter, Azospirillum, Bacillus, Burkholderia, Flavobacterium, Pseudomonas, Rhizobium, etc., play important role in plant growth promotion. In addition, such symbiotic associations of PGPRs in the rhizospheric region also confer protection against several diseases caused by bacterial, fungal and viral pathogens. The biocontrol mechanism utilized by PGPR includes direct and indirect mechanisms direct PGPR mechanisms include the production of antibiotic, siderophore, and hydrolytic enzymes, competition for space and nutrients, and quorum sensing whereas, indirect mechanisms include rhizomicrobiome regulation via. secretion of root exudates, phytostimulation through the release of phytohormones viz., auxin, cytokinin, gibberellic acid, 1-aminocyclopropane-1-carboxylate and induction of systemic resistance through expression of antioxidant defense enzymes viz., phenylalanine ammonia lyase (PAL), peroxidase (PO), polyphenyloxidases (PPO), superoxide dismutase (SOD), chitinase and β-glucanases. For the suppression of plant diseases potent bio inoculants can be developed by modulating the rhizomicrobiome through rhizospheric engineering. In addition, understandings of different strategies to improve PGPR strains, their competence, colonization efficiency, persistence and its future implications should also be taken into consideration.

Concerns have been raised on the effectiveness and sustainability of Soil and Water Conservation (SWC) practices as adaptation options to climate change and high intra− and inter−annual rainfall variabilities in eastern Democratic Republic of Congo (DRC). This study was conducted in the Ruzizi Plain, a dryland area, to assess the performance of maize (Zea mays L.) under two Representative Concentration Pathways (RCP 4.5 and 8.5) and two SWC practices (tied ridges and conventional tillage). The AgMIP’s Regional Integrated Assessment (RIA) approach was used to simulate Water Use Efficiency (WUE) under the Cropping System Model−Crop Environment Resource Synthesis (CSM−CERES−Maize) of the Decision Support System for Agro−technology Transfer (DSSAT). The model was calibrated using experimental data from nine cropping seasons (2011–2018) and 100 farms. The model sensitivity was assessed as a function of temperature, water, and SWC practices for the same environments. Initial conditions of crop management practices were used as input data for CSM−CERES−Maize. Current climate data were extracted from AgMERRA datasets corrected with local data for the period of 1980 to 2021. Future climate projections (2022–2099) were obtained after down−scaling the data from the 29 GCMS of CMIP5 and subsetted to five GCMs based on climate regimes. GCMS results were a strong indicator that climate change in this DRC dryland will result in an increase in average annual temperatures for both RCP 4.5 and 8.5, with the highest increase (3.05 °C) under hot/dry conditions for RCP8.5 and the lowest (1.04 °C) under cool/dry conditions for RCP 4.5. All the models selected for five climate regimes for 2022–2099 showed no change in the rainfall trends for RCP 4.5 (p > 0.05). The models projected yield declines of 5–25%, with less yield losses under tied ridges as an adaptation practice. The use of efficient SWC practices could therefore be a promising strategy in reducing potential losses from climate change in drylands of eastern DRC.
