Background Glioma is characterised by easy invasion of surrounding tissues, high mortality and poor prognosis. Moreover, the prognosis of glioma is getting worse and worse with the increase of grade, which is not optimistic. Therefore, biological markers for glioma are needed in clinical to detect and evaluate the situation and prognosis of patients with glioma. In many studies, we have found that the protein arginine methyltransferase 6 (PRMT6) expression is elevated in various tumors, which is associated with prognosis of patient. However, there has been no report or study on the role of PRMT6 in glioma. Methods In this study, we used various tumor-related databases to analyze the mechanism of PRMT6 in tumors, especially gliomas, from bioinformatics, and carried out relevant experimental verification with tumor tissues extracted from patients during surgery. Besides, we analyzed the relationship between PRMT6 expression and immune infiltration and immune-related cells, and discussed the possible mechanisms. We also discussed the role of PRMT6 expression in glioma from mutation, clinical indicators, enrichment analysis, and immunohistochemical results. Results PRMT6 is significantly differentially expressed in multiple tumors, which is associated with survival and prognosis. Especially in gliomas, the PRMT6 expression gradually increased with the grade increasing. Besides, PRMT6 can be used as an independent prognostic risk factor in the nomogram and has been verified in various databases. Conclusions Our results indicate that high PRMT6 expression is a potential biomarker for predicting prognosis and progression of glioma.

BACKGROUND: Immunotherapy combined with standard of care (SOC) is often used for high-grade glioma (HGG). There are few comparisons of immunotherapy with SOC treatment or without SOC treatment (IMT) versus SOC. It is important to understand what interventions exist and their relative effectivenesses. METHODS: The Cochrane Library, Embase, Medline, and the Web of Science Core Collection were systematically searched by two librarians. Retrieved hits were screened for inclusion. Subgroup analysis was used to examine the main factors associated with overall survival (OS) and progression-free survival (PFS), which were used as primary endpoints to assess the efficacy and safety of IMT. This study was registered with PROSPERO, number CRD42019112356. RESULTS: The search yielded 2,315 results, of which 11 met the eligibility criteria. We identified 6 publications through exploration. Compared to SOC alone, IMT including viral therapy (VT) and dendritic cell vaccination improved the OS (HR = 0.55, 95% CI 0.42-0.72; p < 0.0001) and PFS (HR = 0.67, 95% CI 0.53-0.84; p = 0.0005). CONCLUSIONS: We found that IMT in the era of TMZ has an improved effectiveness over SOC. Our findings support the use of immunotherapy for brain tumors to improve HGG outcomes.

Introdução: O glioblastoma (GBM) é o tumor cerebral primário mais comum e agressivo em adultos. Suas características histopatológicas o apresentam como um processo neoplásico astrocístico grau IV, com importante proliferação microvascular e necrose tumoral. Discussão: Alterações moleculares comuns ao GBM incluem mutações gênicas ocorridas em IDH1/2, p53, Rb, PTEN, EGFR, metilação do promotor MGMT e outros importantes biomarcadores. O tratamento para essa neoplasia refere-se à cirurgia, radioterapia e quimioterapia alquilante. Todavia, mesmo com emprego dessas terapêuticas, o prognóstico dos pacientes é restrito com sobrevida média de 15 meses. Conclusão: As perspectivas atuais concentram-se no desenvolvimento de novas alternativas terapêuticas com alvos, majoritariamente, às características moleculares que impulsionam o fenótipo maligno do tumor.
