The purpose of this preliminary study is to evaluate the push-out bond strength (PBS) of experimental dentin posts (EDPs) obtained from human and bovine teeth sterilized by autoclaving and gamma radiation. Eighty-four mandibulary premolars were obturated and divided into three post groups: the glass fiber group (Fb) human EDP group (Hm) and the bovine EDP group (Bv). Three subgroups (n=12) were obtained for each EDP groups according to the sterilization methods; no sterilization (Cnt), steam autoclaving (Aut), and gamma radiation (Rad) a total dose of 25 kGy. All posts were cemented to root canals using a dual cured resin cement (Panavia SA). After the micro slices (1mm in thickness) were obtained of each subgroup, PBS test was performed. Data were analyzed using two-way ANOVA and Tukey’s multiple comparison tests (α=.05). The post type and sterilization method was significantly effective on the PBS values according to the ANOVA (P<.001). No sterilization apllied EDP groups showed significantly higher PBS values than the sterilized groups (P<.001). Bv_Cnt (9.42 ±1.31) showed significantly lower PBS values than both Fb (12.36 ±1.54) and Hm_Cnt (11.06 ±1.38) groups (P<.001). Both steam autoclaving and gamma radiation affect the PBS values and fracture modes of EDPs negatively. The bovine EDPs are not as effective as human EDPs with regard to the PBS to the root dentin.

Laboratory research in dentistry and dental education use extracted human teeth as a model for simulation for ex vivo procedures. Human-borne tissues are the first choice of material for bond strength assessment. To obtain comparable results in dental material tests and to ensure microbiological safety, specimens must be stored under specific, uniform conditions. The aim of this paper was to present the contemporary view and recommendations on preserving extracted human teeth. The antimicrobial properties of the storage medium are a crucial aspect, as extracted teeth pose a risk of cross-infection. A classification of different methods (using solutions and otherwise) is presented and their sterilizing efficiency is compared based on the literature. The emphasis is put on the interaction between the storage conditions and the substrate. Tooth specimens should be biologically safe and have normal mechanical properties. The sterilizing process must be neutral for the enamel and dentin microstructure, because even a minor change can affect the adhesive bonding. Autoclave sterilization and storage in 10% formalin solution are widespread and reliable methods, although they do have their disadvantages. There is a need for further investigation in order to establish uniform recommendations on preparing and preserving extracted human teeth used for research purposes.

Peracetic Acid: A Practical Alternative to Formalin for Disinfection of Extracted Human Teeth

Extracted human teeth provide the closest approximation to teeth in situ and play important roles in dental education and materials research. Since extracted teeth are potentially infectious, the Centers for Disease Control recommend their sterilization by autoclaving or disinfection by formalin immersion to ensure safe handling. However, autoclaving is not recommended for teeth with amalgam fillings and formalin is hazardous. The goal of the present study was to investigate the potential of peracetic acid (PA) as an alternative method to achieve reliable disinfection of freshly extracted teeth. A total of 80 extracted teeth were collected for this study. Whole teeth were incubated in one of four solutions for defined periods of time: sterile water (2 weeks), formalin (2 weeks), PA 1000 ppm (2 weeks), and PA 2000 ppm (1 week). After sectioning, the crown and root fragments were transferred into separate tubes containing brain–heart infusion broth and incubated at 37 °C under anaerobic conditions for 72 h. Absence of broth turbidity was used to assess effectiveness of disinfection. No turbidity was observed in any of the formalin-treated or peracetic acid-treated samples, signifying complete disinfection. Our results indicate that PA can effectively disinfect extracted human teeth, providing a reliable alternative to formalin and autoclaving.
