Effects of traditional Chinese medicines on pharmacokinetics of levofloxacin

Abstract: The effects of single coadministrations of one of three traditional Chinese medicines, Hotyu-ekki-to, Rikkunshi-to, and Juzen-taiho-to, on the pharmacokinetics of levofloxacin (LVFX) were investigated with eight healthy volunteers in an open, random crossover fashion. Subjects each received a single oral dose of LVFX (200 mg) alone and then with a single coadministration of each Chinese medicine. There were no significant differences in any pharmacokinetic parameters of LVFX between the groups. Also, no signif… Show more

“…The bioavailability of ampicillin, but not amoxicillin, was reduced significantly during a khat (Catha edulis)-chewing session (Fugh-Berman and Ernst, 2001). The pharmacokinetics of a single oral dose of ofloxacin were unaltered by either Sho-saiko-To or Rikkunshi-to (Hasegawa et al, 1995).…”

“…Blood samples (approximately 1 ml) were collected with a vacuum blood collection tube containing EDTA (Venoject II, Terumo, Tokyo, Japan) at time zero (just before the start of administration) and at 0.5, 1, 1.5, 2, 3, 4, 8, 12, and 24 h for ciprofloxacin and at 1,2,3,4,6,8,12, and 24 h for tetracycline after each dose. Blood samples were immediately centrifuged to yield plasma.…”

“…We have previously reported that the Kampo preparations, Hotyu-ekki-to, Rikkunshi-to and Juzen-taiho-to, have no effect on the pharmacokinetics of the quinolone antimicrobial agent levofloxacin in healthy volunteers. 6) To our knowledge, there is no information regarding pharmacokinetic interaction between byakkokaninjinto, which contains a relatively large amount of cationic metals, and antimicrobial agents. However, the therapy with the Kampo preparation is a common practice and is sometimes prescribed without doctors' knowledge of whether or not pharmacokinetic interactions between the Kampo preparation and Western drugs including antimicrobial agents occur.…”

“…Many herb-Western medicines or Kampo medicinesWestern medicines interactions have been discussed before [2][3][4][5][6]. Coadministration of St. John's wort, a herb extensively used as an antidepressant [7], potently inhibited the activities of major human drug-metabolizing enzymes [8].…”
