Shoot proliferation is a very important micropropagation phase, decisive for economic efficiency of this method for a given taxon. To obtain a high multiplication ratio and a good quality of microshoots a detailed propagation protocol must be developed for particular species or even cultivars. Rhododendron ‘Kazimierz Odnowiciel’ is a relatively new cultivar distinguished by large, beautiful flowers and high frost resistance so there is a need to develop an efficient method of its propagation to satisfy a growing demand for this plant. The aim of the experiment was to evaluate effects of cytokinins: meta-Topolin (mT), zeatin (ZEA), 6-benzyladenine (BA), thidiazuron (TDZ), 2-isopentenyladenine (2iP), or the combination of 2iP+ZEA on proliferation of shoots in R. ‘Kazimierz Odnowiciel’ cultured on Anderson’s medium (AN). Biochemical changes in plant material affected by cytokinins during this phase of micropropagation were determined and occurrence of genetical changes was followed using ISSR markers. TDZ, ZEA or the combination of ZEA+2iP resulted in 100% explant regeneration. On the medium with TDZ or ZEA over two new shoots per explant were produced but the highest proliferation was attained on the medium containing ZEA+2iP – over three shoots per explant. Microshoots developed in this treatment had also the highest contents of chlorophyll, carotenoids and soluble sugars as well as the highest catalase activity. Microshoots formed on the medium with zeatin showed the lowest polymorphism (below 4%) relative to a stock plant.

The objective of this study was to evaluate the effect of auxin pulse intervals on the induction of somatic embryos of Eucalyptus grandis x E. urophylla and to describe the embryogenic behavior of callus under the effect of auxinic stress. Cotyledons were inoculated in culture medium containing 207.07 µM picloram, a treatment considered as auxin pulse. Explants that were in the auxin pulse treatment were transferred to semisolid or liquid medium containing 20.71 µM picloram after one, two, four or eight days of auxin pulse. In a second experiment, explants that were on auxin pulse treatment were transferred to semi-solid medium containing 20.71 µM picloram after one, two or three days of auxin pulse. Auxiliary picloram pulse treatments (207.02 µM) can be used as an initial source of stress for the acquisition of embryogenic competence. The oxidation of cotyledonary explants may be considered as an indication of the formation of embryogenic calli. The presence of pectins in peripheral regions of somatic pro-embryos can be considered as a marker of somatic embryogenesis in cotyledonary explants of Eucalyptus grandis x E. urophylla.

In this modern era, farmers are adopting high density planting (HDP) for most of the fruit crops. Dwarfing is based on the principle of making the optimal use of vertical and horizontal space per unit time and harnessing the highest potential returns per unit of inputs and natural resources. Dwarfing in fruit crops can be accomplished through a variety of methods, including the use of dwarfing rootstock, the selection of spur type scion cultivars, use of interstock, pruning, root pruning, various training systems, girdling, scoring, bark invasion and the application of growth retardants. One strategy for promoting early fruiting, less vigour and increasing production is to utilize dwarf root stocks. The mechanism underlying dwarfing involves anatomical, physiological and biochemical changes. It has been proposed that dwarfing rootstock or interstock control tree size by regulating the auxin that passes through the rootstock or interstock’s bark. Paclobutrazol retards growth, shoot elongation and reduced internodal length by inhibiting gibberellins synthesis. Dwarfness can be imparted by the using incompatible rootstock. Dwarfing can be induced by inducing viral infection, although, it is not commercially adopted. Recombinant DNA technology has recently expanded the gene pool that can be managed to cause dwarfism and maximize benefits in horticulture crops.
