The protective and/or therapeutic effect of berberine on cyclophosphamide-induced testicular injury was investigated in rats. A total of 40 male Sprague-Dawley rats were divided into 5 groups (n=8 per group). The control group received saline and CP, BER75, BER150 and BER300 groups received single dose of intraperitoneal 200 mg/kg CP on day 8. Berberine (75 mg/kg, 150 mg/kg and 300 mg/kg) was administered orally for 7 days before and after the CP injection. At the end of the protocol, testes were removed for histological examination, immunohistochemical staining, and biochemical assessments. In our study, CP did not cause a significant change in body weight and testis weight, whereas berberine treatment significantly decreased body weight. Although no significant change was observed in terms of oxidative stress markers and cytokine levels, degenerative changes and apoptotic findings were found in the CP group; which were improved significantly with increasing doses of berberine treatment. In conclusion, we found that treatment with berberine may have anti-degenerative and anti-apoptotic potential rather than altering oxidative stress markers and/or inflammatory pathways in CP-induced testicular damage.

Background: Cyclophosphamide is a commonly used anticancer and immunosuppressive agent; however, hepatotoxicity is one of its severe toxicities. Hydrogen sulfide is a gaseous signaling molecule that plays crucial regulatory roles in various physiological functions. This study aimed to evaluate the hepatoprotective effect of hydrogen sulfide against cyclophosphamide-induced hepatic damage in rats. Methods: Hepatotoxicity was induced by the single intraperitoneal administration of cyclophosphamide (200 mg/kg). Sprague–Dawley rats were treated by hydrogen sulfide donor, sodium hydrosulfide (25, 50, and 100 µmol/kg, intraperitoneal) 7 days before and 7 days after the administration of a single intraperitoneal injection of cyclophosphamide (200 mg/kg). Cyclophosphamide-induced hepatotoxicity was evaluated by serum and tissue biochemical and histopathological assessments. The levels of hydrogen sulfide, nitric oxide, cyclic guanosine monophosphate, interleukin 6, and interleukin 10 in liver homogenates were also determined by ELISA. One-way analysis of variance and Kruskal–Wallis tests were used as statistical analyses. Results: Cyclophosphamide increased liver function enzymes (alanine aminotransferase and aspartate aminotransferase), immunoreactivity to caspase-3 and Apaf-1, and proinflammatory cytokines. Cyclophosphamide also induced histopathological alterations including pycnotic nucleus with eosinophilic cytoplasm, increased sinusoidal dilatation, congestion, and edema. Hydrogen sulfide co-treatment significantly reduced cyclophosphamide-induced inflammation, histological alterations, and apoptosis in the liver. 50 mg/kg sodium hydrosulfide was more effective against cyclophosphamide-induced hepatotoxicity. Conclusion: In conclusion, hydrogen sulfide with its anti-inflammatory and anti-apoptotic effects seems to be beneficial as an adjunct to cyclophosphamide treatment to reduce cyclophosphamide-induced hepatotoxicity and thereby can be suggested as a promising agent to increase the therapeutic efficacy of cyclophosphamide.

Androgens are essential for male physical activity and normal erectile function. Moreover, estrogens also influence erectile function, and high estrogen levels are a risk factor for erectile dysfunction (ED). In this review, we summarize relevant research examining the effects of the sex hormone milieu on erectile function. Testosterone affects several organs, particularly erectile tissue. The mechanisms through which testosterone deficiency affects erectile function and the results of testosterone replacement therapy have been extensively studied. Estrogen, the female sexual hormone, also affects erectile function, as demonstrated in both clinical and basic studies. Interestingly, estradiol-testosterone imbalance is considered a risk factor for ED. Furthermore, endocrine-disrupting chemicals have estrogen-like effects and cause ED. Phosphodiesterase-5 (PDE-5) inhibitors, first-line drugs for the treatment of ED, increase the levels of testosterone and estradiol in patients with low testosterone levels. Therefore, estrogen levels should be carefully monitored in patients receiving PDE-5 inhibitors. Future studies are needed to confirm these findings using molecular tools in order to provide insights into the treatment and mechanisms of endocrine-related ED.
