This study was conducted to develop long-term storage methods for turkey semen using different extenders. During the study, the massage method was used twice a week to collect the semen from five turkeys, a total of 44 times. The collected fresh semen's average ejaculate quantity, sperm concentration, motility and vitality values were determined as 0.22±0.01 ml, 3.5±0.17 x10 9 sp/ml, 77.0±1.44% and 86.2±0.95 % respectively. In this study, glucose (G) including 5% Dimethyl sulphoxide (DMSO) cryoprotective, tris-glucose (TG), lactated Ringer's (LR), and lactated Ringer's glucose (LRG) extenders were used. The Turkey semen was combined and divided into four equal parts, and subsequently diluted at a ratio of 1:3 and equilibrated at +4°C for 90 minutes. Following equilibration, the samples were frozen in liquid nitrogen vapor to -80°C for five minutes, and stored at -196°C in liquid nitrogen. After freezing and thawing, the highest motility value was obtained from the G extender (43.3% ±1.62%) followed by LRG (24.6±1.53%), LR (12.6±0.92%) and TG (12.2±0.66%).The vitality values were recorded as 55.8±1.89%, 23.8±1.58%, 21.5±1.10% and 36.5±1.59% respectively. The motility and vitality values were significantly (p<0.01) more for the glucose extender than those for other extenders. Therefore, it was concluded that glucose extender is better option for the long-term storage of Turkey semen.

Semen from individual roosters was collected in a 1.5 mL microtube containing 0.1 mL of IGGKph diluent, which was composed of 0.14 g potassium citrate, 1.40 g sodium glutamate, 0.21 g sodium dihydrogen phosphate, 0.02 g protamine sulfate, 0.98 g anhydrous sodium hydrogen phosphate, 0.70 g glucose, 0.02 g fructose, 0.70 g inositol, and 0.10 polyvinylpyrrolidone in 100 mL distilled water [ 19 ]. The effect of age on fertility was investigated in Thai native chickens. The objective of this study was to determine the effects of age (mature and old) on the morphological characteristics of the reproductive organs and the histological characteristics of the uterovaginal junction (UVJ) tissues, resident sperm in the UVJ, and fertility duration in Thai native hens. We found no differences in the morphological characteristics of the reproductive organs, except for the number of follicles and the sizes of the fifth large yellow follicle in mature hens, which were greater than those in old hens (p < 0.05). The diameter of the sperm storage tubules (SSTs) epithelium was larger in old hens than in mature hens (p < 0.05), whereas the epithelium height was lower in old hens (p < 0.05). The number of sperm in the SSTs was greater in mature hens compared with old hens (p < 0.05). Mature hens showed a higher fertility rate than old hens. Our results suggest that, in old hens, the function of the SSTs is impaired, and sperm cannot be retained. Such a deterioration of the SSTs may be one of the factors involved in the decline in fertility.

The aim was to estimate the optimal frequency of semen collection from pigeons in relation to ejaculate volume, sperm concentration, total spermatozoa in ejaculate and percentage of live morphologically normal cells. The study was carried out on 455 ejaculates collected from two groups of pigeons, each of 10 males (group I: meat-type breed; group II: fancy pigeon). The birds were selected and kept individually in cages under a natural photoperiod. A two-person technique was used for semen collection (lumbo-sacral and cloacal region massage). Semen was collected once, twice or three times per week. Colour, consistency and volume of ejaculates were evaluated macroscopically immediately after collection. Sperm concentration and total number of cells in the ejaculate were estimated after dilution with Ringer's solution. A live-dead stain technique (nigrosin-eosin) was used to determine the percentage of live and normal spermatozoa. Semen collected 3x/week was of high quality. The average volume of a single ejaculate was small (21 microl in group I and 19 microl in group II), but sperm concentration was high--1.58 x 10(9)/ml and 1.96 x 10(9)/ml, respectively. The mean number of spermatozoa per ejaculate was 30.48 x 10(6) in group I and 39.49 x 10(6) in group II. An increased percentage of live and normal spermatozoa in semen collected more frequently was also observed. Collecting pigeon semen 3x/week provides spermatozoa in larger amounts and of better quality than less frequent collections (1x/week or 2x/week) and is recommended for obtaining more insemination doses.
