Effects of Progesterone on iNOS, COX-2, and Collagen Expression in the Cervix

Abstract: S U M M A R Y This study examines the relationship between inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in the control of cervical ripening and parturition under normal (normal term pregnancy) and abnormal (preterm labor and prolongation of pregnancy) conditions by (a) measuring changes in the collagen both visually and quantitatively, (b) localizing and characterizing iNOS and COX-2 under normal conditions, and (c) characterizing the changes in iNOS and COX-2 under abnormal conditions. … Show more

“…Indeed, the experimental findings suggested that the mechanism of progesterone involved an interaction between iNOS and P38MAPK signal pathway [23] . The impact of progesterone on the MAPK activity to suppress NO, which we concerned, were consistent with previous studies [24] .…”

“…COX-2, resulting in transcriptional activation that promote uterine contractility, is important during gestation and parturition (6,7,22,23). The COX-2 mRNA levels in our study appeared to be compatible with the several studies reported in literature (3,7,31,15,28). According to our results, COX-2 mRNA levels gradually increase at the beginning of pregnancy from day 10 to 20 and expression of COX-2 was at a maximum on day 21 (parturition).…”

“…Studies show that iNOS and COX-2 enzymes were co-localized during gestation under normal conditions (25,30). Marx et al also demonstrated the relationship between COX-2 and iNOS in the control of cervical ripening and parturition under both term and preterm laboring conditions (15). Moreover, recent studies reported that iNOS mRNA levels had a tendency to decrease whereas COX-2 mRNA levels increased as pregnancy progressed to term, demonstrating an inverse relationship (7,8,24).…”

“…MMP-2 and MMP-9 in the cervical ripening process has been indicated in cervical ripening. 6,7 To summarize, the complex interactions of various cytokines bring about profound changes in the proteoglycans in the cervix which eventually leads to cervical ripening.…”
