EFFECTS OF pH AND OF VARIOUS CONCENTRATIONS OF SODIUM, POTASSIUM, AND CALCIUM CHLORIDE ON MUSCULAR ACTIVITY OF THE ISOLATED CROP OF PERIPLANETA AMERICANA (ORTHOPTERA)

Abstract: In 1940, Griffiths and Tauber (13) published a preliminary account of attempts to devise a physiological salt solution for the isolated foregut of the American roach, Periplaneta americana L. Initially, they tested insect physiological solutions which were recommended by B~l~r (2), Hobson (16), Yeager (25), TenCate (23), and Glaser (10). Only the solutions of l~l ' r and Hobson produced results which were at all satisfactory for the excised crop of the American roach. Secondly, amounts ~ of the chlorides of so… Show more

“…Organic acid production by foregut homogenates. Intact foreguts were dissected out of five P. americana adults and pooled in a small glass tissue homogenizer containing 4.5 ml of Ringer solution, pH 7.8 (19). Foreguts were homogenized for 1 min, and 1-ml portions of the homogenate were added to 5-ml glass serum vials.…”

“…Colons were dissected out, slit lengthwise, and vortexed in insect Ringer solution (8) to remove the lumen contents. They were then fixed by a modified osmium tetroxide-ANTIMICROB.…”

“…After :::::2 h of feeding on dual-labelled leaf disks, insects were anesthetized in C0 2 -saturated water and carefully dissected. Exposed guts were bathed in a saline solution (Griffiths and Tauber 1943) while the ends were double ligated with polyester thread. The ligated guts were excised and incubated in 4 mL of saline solution for 3 hat 15.…”
