Effects of paeonol on intracellular calcium concentration and expression of RUNX3 in LoVo human colon cancer cells

Abstract: Paeonol, a major phenolic component of the root bark of Paeonia moutan, is known to exhibit antitumor effects. However, the underlying mechanisms remain unknown. In the present study, the effects of paeonol on cell viability, intracellular calcium concentration and the expression of runt‑related transcription factor 3 (RUNX3) were analyzed in LoVo human colon cancer cells. Results revealed that paeonol markedly reduced LoVo cell viability in a time‑ and dose‑dependent manner. Flow cytometry assays demonstrated… Show more

“…37,38 In human colon cancer LoVo cells, paeonol blocked cell cycle at the G1 to S transition and induced apoptosis. 39 In HT-29 cells, paeonol inhibited proliferation 40 and showed a synergetic antiproliferative effect when combined with 5-FU. 41 In human esophageal adenocarcinoma cells, paeonol showed a dose-dependent growth-inhibitory effect, and this effect was synergistic when combined with cisplatin.…”

“…1) [2]. Paeonol exhibits antibacterial, antioxidant, anti-aggregation, anxiolytic-like, and anti-inflammatory activities [3][4][5][6][7][8][9]. Therefore, paeonol content has been used as a chemical index for the quality of crude drugs.…”

“…Paeonol, the primary phenolic component in Radix Paeoniae Alba (Shaoyao), reduces HT29 cell proliferation by downregulating cyclooxygenase-2 and upregulating p27 [10]. Paeonol induces LoVo cell apoptosis by increasing intracellular calcium concentration [11]. However, whether or not SYD can prevent colitis-associated CRC (caCRC) remains elusive.…”
