Effects of Medicinal Fungi-Derived β-Glucan on Tumor Progression

Abstract: β-Glucans have been studied in animal species, from earthworms to humans. They form a heterogenous group of glucose polymers found in fungi, plants, bacteria, and seaweed. β-Glucans have slowly emerged as an important target for the recognition of pathogens. In the current review, we highlight the major roles of mushroom-derived β-glucans on cancer progression.

“…The adjuvant mechanism of immune system activation depends on the formation of an immune complex of β-glucans, after recognition as PAMP, with anti-β-glucan antibodies, which through immune effector cells and C3 phagocytes facilitates killing of tumor cells, as represented in Figure 2 [37]. The major roles of mushroom-derived β-glucans on cancer progression have been extensively reviewed [38]. A promising application of β-glucans is their use in cancer treatment, as adjuvant of conventional therapies.…”

“…Bioactive compounds, such as -glucans, were found in these L. rhinocerus MB-EPS extracts; use of glucans have gained appeal in recent years for a number of emerging applications including biopolymers 30 and biomedicines 31 . It is notable that mushroom-derived -glucans have many new therapeutic properties, including (a) potential new or complementary immunotherapies against Coronavirus disease (SARS-CoV-2) 14 , (b) potential new therapeutic agent for mitigating diseases associated with gastrointestinal mucosal damage, such as peptic ulcers and inflammatory bowel disease 32 , (c) potential as anticancer drugs for lung and breast cancer 33 , and (d) potentially asthmatic treatment 23,34 . However, there is a marked gap in knowledge surrounding the toxicity (if any), of these mushroom-derived bioactive compounds; particularly use of Zebrafish trials that would aid product development and implementation.…”

“…β−glucans linked with chitin chains ensure the simultaneous rigidity and flexibility of the fungal cell wall. These compounds most often have the structure of β-D-glucans or β-mannans [ 100 , 101 ]. Their activity is determined by their molecular weight, solubility in water, number of branches and tertiary structure.…”
