Effects of Medical Marijuana on Migraine Headache Frequency in an Adult Population

Abstract: The frequency of migraine headache was decreased with medical marijuana use. Prospective studies should be conducted to explore a cause-and-effect relationship and the use of different strains, formulations, and doses of marijuana to better understand the effects of medical marijuana on migraine headache treatment and prophylaxis.

“…The medical literature regarding treatment of headache, migraine, and facial pain disorders shows supporting evidence for cannabis/cannabinoids in the treatment of chronic headaches, migraine including chronic migraine, medication overuse headache, cluster headache, idiopathic intracranial hypertension, and multiple sclerosis (MS) associated trigeminal neuralgia . At the time of this writing, this majority of supporting literature consists primarily of case series, case studies, case reports, surveys, clinical/anecdotal reports, and one retrospective analysis.…”

“…It would be interesting to explore whether migraine, anxiety and sleep disorders in patients with rare deleterious variants in CNR1 could be alleviated by treatment with THC or other CB1 agonists that may effectively stimulate the impaired and/or remaining functional CB1 receptors. In this regard, we note that a recent observational study of 121 adults with migraine reported a decrease in mean headache frequency from 10.4 to 4.6 headaches per month (p<0.0001) for patients receiving medicinal cannabis [71].…”

“…These studies suggest the efficacy and interactions of cannabis or THC with the eCB may be dependent on usage, as well as final potency of the CBs. A recent retrospective study found medical marijuana to effectively reduce the frequency of headaches in patients, however, various side-effects were reported depending on the type of marijuana [109]. In regard to migraine pain, THC stimulates 5-HT synthesis [110], is a 5HT1A agonist, and 5HT3 antagonist implicating its role in the treatment of migraine and pain with several other potential mechanisms of action [111].…”
