Effects of mechanical uncouplers, diacetyl monoxime, and cytochalasin-D on the electrophysiology of perfused mouse hearts

Abstract: concentration ([Ca 2ϩ ]i) handling and vulnerability to arrhythmias in a species-dependent manner. The effects of uncouplers were investigated in perfused mouse hearts labeled with rhod-2/AM or 4-[␤-[2-(di-n-butylamino)-6-naphthyl]vinyl]pyridinium (di-4-ANEPPS) to map [Ca 2ϩ ]i transients (emission wavelength ϭ 585 Ϯ 20 nm) and action potentials (APs) (emission wavelength Ͼ 610 nm; excitation wavelength ϭ 530 Ϯ 20 nm). Confocal images showed that rhod-2 is primarily in the cytosol. DAM (15 mM) and cyto-D (… Show more

“…The application of blebbistatin induced a transient increase in cycle length. However, blebbistatin did not induce any shifts in leading pacemaker sites or changes in action potential duration, as has been associated with other contraction blockers (2,26). This is consistent with what was observed in our previous studies using human tissue (11) and animal models (dog, rabbit, and rat) (13,14,17).…”

“…Other popular heart immobilizers (butanedione monoxime and cytochalasin D) were shown to have various effects on cardiac electrophysiology, arrhythmias, and energy balance in normal or ischemic hearts (3,21). Complete exclusion of immobilizers from physiological cardiac imaging is difficult.…”

“…Hearts were perfused and suspended in a customdesigned optical chamber (32). Di-4-ANNEPS and Rhod2 loading, photodiode array mapping, restitution protocols, and analysis were as described (40). GCaMP2 was excited at 488 nm, and di-4-ANEPPS and Rhod-2 were excited at 530 nm.…”
