Effects of mannosylated glycopolymers on specific interaction to bone marrow hematopoietic and progenitor cells derived from murine species

Abstract: Poly[N-pvinylbenzyl-O-D-galactopyranosyl-(1-4)-D-glucoamide], poly[N-pvinylbenzyl-O-D-glucopyranosyl-(1-4)-D-glucoamide], and poly[N-p-vinylbenzyl-O-mannopyranosyl-(1-4)-D-gluconamide] (referred to as PVLA, PVMA, and PV-Man) are polystyrene derivatives that contain galactose, glucose, and mannose moieties, which interact with hematopoietic cells (HCs). To clarify the specific interaction between the glucopolymers and hematopoietic cells, glycopolymers labeled with fluorescent isothiocyanate (FITC) were used to… Show more

“…20 In this study, we confirm the possibility of the binding activity of polymers with human hematopoietic cells derived from CB and PB. Using FITClabeled glycopolymers, we tested the dose dependent binding effects of polymers with human hematopoietic cells from CB and PBL.…”

“…In a previous report, PV Mannose showed higher binding capacity than PV Maltose and PV Lactose glycopolymers to murine hematopoietic cells. 20 Therefore, human and murine hematopoietic cells show a similar glycopolymer binding profile.…”

“…Mac1 has been shown to bind type-I fimbriated E. coli in a mannose dependent manner 53 and CD11b+ monocytes shows high binding affinity to mannosylated glycopolymers 54 . Soluble CD11b demonstrates elevated binding to ICAM-1 produced in the presence of alpha-mannosidase 1 inhibitors 55 suggesting that increased mannose on ICAM-1 promotes binding.…”

“…The low water solubility of Cel-O-Lac probably arises from its hydrophobic -face of non-reducing -galactoside units. The observed negative effects arising from the hydrophobic spacers and the -face on their water solubility are much more drastic in comparison with those observed for glycoclusters, based on flexible artificial polymers such as polystyrene [34][35][36][37][38][39][40] and polyacrylamide [41][42][43][44][45][46]. For example, polystyrenes having -lactoside appendages still have excellent water solubility.…”
