Effects of Hypericum perforatum extract on oxaliplatin-induced neurotoxicity: in vitro evaluations

Abstract: Hypericum perforatum L. has been used for centuries as a natural remedy for the treatment of many disorders. Neuropathic pain is a common side effect of oxaliplatin-based chemotherapy and often the cause of therapy discontinuation. Thanks to its anti-inflammatory and analgesic effects, the use of H. perforatum may be a novel therapeutic strategy for neuropathy. The aim of this paper was to evaluate the effect of H. perforatum hydrophilic extract on an in vitro model of oxaliplatin-induced neurotoxicity. The an… Show more

“…Common antitumor drugs such as oxaliplatin are reliable for the clinical treatment of gastric cancer. However, long-term use of oxaliplatin can result in nephrotoxicity [20], ototoxicity [21], neurotoxicity [22], and myelosuppression [23]. Moreover, the resistance of tumor cells to oxaliplatin reduces the efficacy of the drug and is a major cause of chemotherapy failure [24].…”

“…Hypericum perforatum (St. John's Wort) is used for centuries as a natural remedy for the treatment of a variety of disorders including depression (Asgary et al, 2012 ; Abtahi Froushani et al, 2015 ). It can reduce oxaliplatin-induced caspase-3 activity in rat astrocytes, but its alone produces a cytotoxic effect and fails to reduce the cytotoxicity of oxaliplatin in HT-29 cancer cells (Cinci et al, 2017 ). Its main constituent hypericin is a potent inhibitor of glioma growth in vitro .…”

“…Hypericum perforatum L. (St. John’s wort) is well-known for its antioxidant, anti-inflammatory, analgesic, and neuroprotective effects [ 151 , 152 , 153 ]. Its extract showed significant protective activity against oxaliplatin-induced neurotoxicity in vitro and reduced caspase-3 activity in rat astrocytes without the reduction of oxaliplatin cytotoxicity on HT-29 cells [ 135 ]. Satureja hortensis aerial part extract, applied in rats at three different concentrations (50, 100 and 200 mg/kg) along with cisplatin injection, exhibited strong anxiolytic activity and lowered apoptotic parameters in rat hippocampal tissues [ 127 ].…”
