Effects ofAngelica gigasNakai as an Anti-Inflammatory Agent inIn VitroandIn VivoAtopic Dermatitis Models

Abstract: We investigated the cellular and molecular mechanisms mediating the effects of Angelica gigas Nakai extract (AGNE) through the mitogen-activated protein kinases (MAPKs)/NF-κB pathway using in vitro and in vivo atopic dermatitis (AD) models. We examined the effects of AGNE on the expression of proinflammatory cytokines and chemokines in human mast cell line-1 (HMC-1) cells. Compound 48/80-induced pruritus and 2,4-dinitrochlorobenzene- (DNCB-) induced AD-like skin lesion mouse models were also used to investigat… Show more

“…Since a large number of herbal medicines and their secondary metabolites display anti-inflammatory effects [12,13,28,31], they have been traditionally used as remedies and currently utilized as complementary medicines [31,32]. Among medicinal plants, the AGN extract was verified to have excellent anti-inflammatory effects.…”

“…Among medicinal plants, the AGN extract was verified to have excellent anti-inflammatory effects. The ethanol extract of AGN, for instance, displayed anti-inflammatory effects by reducing the production of histamine and atopy-related cytokines and suppressing the levels of COX-2, NF-κB, and Iκ-Bα in both in vivo and in vitro models of atopic dermatitis [12]. Additionally, AGN extract prevented epithelial injury in colonic tissues of a murine model of dextran sulfate sodium-induced ulcerative colitis by moderating the activation of inflammatory mediators, such as IL-6, TNF-α, prostaglandin E2 (PGE2), and cyclooxygenase-2 (COX-2) [33].…”

“…We retrieved the chemical compounds present in the four herbal medicines that constitute SMT (i.e., AGR, RRP, PRA, and CR) from traditional Chinese medicine (TCM)-related databases, including the Traditional Chinese Medicine Systems Pharmacology (TCMSP) database, Traditional Chinese Medicine Integrated Database (TCMID), and Herb Ingredients' Targets (HIT) database [49][50][51][52]. Regarding AGR, we first explored the biochemical constituents of Angelicae Sinensis Radix, a commonly used herbal medicine in China that belongs to the Angelicae species, using the aforementioned TCM-related databases, and combined those with the reported major compounds of AGR such as marmesin, lomatin, and decursin [53][54][55]. Chemical compounds not contained in AGR, including isoeugenol, stigmasterol, and 4-octanone, were excluded from the integrated data based on previous studies [53][54][55].…”
