Effects of human collagen α-1 type I-derived proteins on collagen synthesis and elastin production in human dermal fibroblasts

Abstract: Collagen type I is the most abundant form of collagen in human tissues, and is composed of two identical α-1 type I chains and an α-2 type I chain organized in a triple helical structure. A previous study has shown that human collagen α-2 type I (hCOL1A2) promotes collagen synthesis, wound healing, and elastin production in normal human dermal fibroblasts (HDFs). However, the biological effects of human collagen α-1 type I (hCOL1A1) on various skin properties have not been investigated. Here, we isolate and id… Show more

“…The TGF-β1 stimulated increase of both gene and protein levels of type I collagen and fibronectin in both fibroblast types and of type III collagen in pulmonary fibroblasts correlates with previous findings in SSc, fibrotic pulmonary, and healthy dermal and pulmonary fibroblasts 22 26 , 28 , 29 , 45 , 46 . The increase of FN1 and FBN-C, PRO-C1 and PRO-C6 in both fibroblast types, and PRO-C3 in dermal fibroblasts by PDGF-AB stimulation is consistent with previous findings 28 , 41 , 47 .…”

“…There are various types of collagen, and the dermis is mainly composed of 80%–85% collagen type I and 10%–15% collagen type III. Collagen type I is a trimer mainly composed of two α1 chains and one α2 chain, encoded by the COL1A1 and COL1A2 genes, respectively 14,15 . Both Col1a1 and Col1a2 mRNA expression levels were significantly decreased in the skin of the SAMP mice compared with the SAMR mice (Fig.…”

“…The reported results show that collagen hydrolysate, derived from bovine type I collagen, notably modulated cell metabolism, increasing the content of type I procollagen and collagen by stimulating their biosynthesis and inhibiting MMP-1 and MMP-2 on both chronological aged and photoaged HDF. In Hwang et al's [51,52] studies, human alpha-1 and alpha-2 type I collagen (COL1A1 and COL1A2)-derived peptides revealed stimulatory effects on type I collagen and elastin production on HDF. Both COL1A1 and COL1A2-derived peptides exerted the same effect promoting collagen synthesis and elastin production.…”
