To eliminate the microbial infection from an injury site, various modalities have been developed such as dressings and human skin substitutes. However, the high amount of reactive oxygen species, microbial infection, and damaging extracellular matrix remain as the main challenges for the wound healing process. In this study, for the first time, green synthesized silver nanoparticles (AgNPs) using Teucrium polium extract were embedded in poly lactic acid/poly ethylene glycol (PLA/PEG) film to provide absorbable wound dressing, with antioxidant and antibacterial features. The physicochemical analysis demonstrated, production of AgNPs with size approximately 32.2 nm and confirmed the presence of phytoconstituents on their surface. The antibacterial assessments exhibited a concentration-dependent sensitivity of Staphylococcus aureus and Pseudomonas aeruginosa toward biosynthesized AgNPs, which showed a suitable safety profile in human macrophage cells. Furthermore, oxidant scavenging assays demonstrated exploitation of plant extract as a reducing agent, endows antioxidant activity to biogenic AgNPs. The formation of PLA/PEG nanofilm and entrapment of AgNPs into their matrix were clearly confirmed by scanning electron microscopy. More importantly, antibacterial examination demonstrated that the introduction of biogenic AgNPs into PLA/PEG nanofibers led to complete growth inhibition of P. aeruginosa and S. aureus. In summary, the simultaneous antioxidant activity and antimicrobial activity of the novel biogenic AgNPs/PLA/PEG nanofilm showed its potential for application as wound dressing.activities of porous Ag nanofilm were investigated. We anticipate that the Ag nanofilm wound dressing with free radical scavenging capacity would enhance the wound healing process. Moreover, this study proposed a low-cost and facile method for preparation of Ag nanofilm as a potential wound dressing. Materials and MethodsMaterials. Methanol (CH 3 OH, 99.9%), silver nitrate (AgNO 3 ), nutrient agar, ferric chloride hexahydrate (FeCl 3 , 6H 2 O), iron (II) sulfate tetrahydrate (FeSO 4 .4H 2 O), dichloromethane, polyethylene glycol 300, ascorbic acid and Mueller-Hinton agar (MHA) were purchased from Merck (Germany). 2,2-diphenyl-1-picrylhydrazyl (DPPH), 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) and 2,4,6-Tripyridyl-s-Triazine (TPTZ) were purchased from Sigma (Germany) and poly lactic acid (PLA) from Zhejiang Hisun Biomaterials (China). The RAW264 macrophage cell line was purchased from National Cell Bank, Pasteur Institute of Iran, Iran and Dulbecco's Modified Eagle's Medium (DMEM), Fetal Bovine Serum (FBS), 1% Penicillin-Streptomycin from Gibco (Paisley, UK). Attenuated total reflection Fourier-transform infrared (ATR-FTIR) spectroscopy is a surface-sensitive and label-free technique, which is applied to obtain dynamic structural information of biomolecules. The study of proteins by ATR-FTIR spectroscopy can be impeded by their tendency to adsorb to solid surfaces. Furthermore, the adsorption process of proteins is often accompanied with conformational changes, which can interfere with the intended structural analysis. We efficiently modified a silicon ATR crystal surface with polyethylene glycol and thereby create a protein-repellent surface. To achieve a high sensitivity, which enables the study of small conformational changes of biomolecules, we combine surface passivation with specific immobilization. This is accomplished via the biotin-streptavidin interaction, which is one of the strongest known non-covalent protein-ligand interactions. As a proof of concept we present the specific immobilization of DNA. The modified surface is stable against elevated temperatures and 8 M urea and can therefore be used to study a wide range of biochemical systems and reactions. The surface chemistry is simple and performed under mild conditions, which leads to a high applicability of the presented approach.

The objective of this study is to characterize the effect of in situ heat treatment on polylactic acid (PLA) and plasticized PLA during fused deposition modelling (FDM) with the motivation to improve their mechanical, thermal and physical properties. The in situ heat treatment was formed during the FDM by adjusting the bed temperature to 70°C, 90°C and 120°C. The performance of 3D, the printed samples, was compared with the compression moulded samples treated at the same temperature using a vacuum oven. PLA was plasticized with poly (ethylene glycol) (PEG) at different compositions of 0, 5 and 10 wt% of PEG. The properties of PLA, plasticized PLA and FDM-printed sample were analysed using Instron, differential scanning calorimeter (DSC) and X-ray diffraction (XRD). It was found that the addition of PEG into PLA decreased the tensile strength, elongation at break and tensile modulus of the materials. However, after heat treatment in the vacuum oven and FDM, the properties were generally higher at 90°C and this was believed to be primarily due to the effect of crystallization. This can be proved by formation of double melting peak, correspond to melt-crystallization mechanism, observed in DSC. The formation of different crystal was supported by XRD analysis where the amorphous peak had transformed into sharp peak at 16.9° and 19.3°, which indicates an improved crystallinity. Comparison between the compression moulded sample and FDM demonstrated that the in situ heat treatment in FDM had the most significant impact on tensile modulus.
