Effects of Garlic on 7,12-Dimethylbenz[a]anthracene-Induced Hamster Buccal Pouch Carcinogenesis

Abstract: The inhibitory effect of garlic (Allium sativum Linn) on 7, 12-dimethylbenz[a]anthracene (DMBA)-induced buccal pouch carcinogenesis was investigated in male Syrian hamsters. Measurement of lipid peroxidation, reduced glutathione (GSH), glutathione peroxidase (GPx), and glutathione S-transferase (GST) was used to monitor the chemopreventive potential of garlic. All hamsters painted on their buccal pouches with DMBA alone for 14 weeks developed well-differentiated squamous cell carcinomas. Diminished lipid perox… Show more

“…Recently, we have reported dose-dependent protection of tomato paste against DMBA-induced genotoxicity and oxidative stress in mice as well as the incidence of hamster buccal pouch tumors (4,9). The antigenotoxic and anticarcinogenic effects of garlic and its organosulfur constituent SAC have also been reported by us as well as by others (3,5,9).…”

“…Epidemiological studies have provided evidence that an increased intake of tomato and garlic is associated with a decreased cancer risk (2,3). In previous studies from this laboratory, we demonstrated the modulatory effects of tomato and garlic on lipid peroxidation, antioxidant and detoxification systems during chemoprevention of 7,12-dimethylbenz[a]anthracene (DMBA)-induced oral carcinogenesis (4,5).…”

“…Balasenthil and coworkers have investigated the effect of aqueous extract of garlic on lipid peroxidation and levels of antioxidants during buccal pouch carcinogenesis in hamsters. They observed that the administration of AGE diminished lipid peroxidation in oral tumor tissue and increased GSH level and glutathione peroxidase activity [Balasenthil et al, 1999]. AGE inhibited LDL oxidation and minimized endothelial cell injury by preventing depletion of intracellular GSH and by removing peroxides [Ide and Lau, 1999].…”

“…Alliin and one of its metabolites, allicin, have also been found to show tumor-inhibiting effects (Lawson, 1998;Dorant, 1993). Since then, although a number of studies have been made on the antitumor and cytotoxic actions of garlic and its organosulfur constituents (Hayes et al, 1987;Wargovich et al, 1988;Balasenthil, 1999;Siegers et al, 1999;Hong et al, 2000;Lamm et al, 2000) the precise mechanisms of the anticarcinogenic action of garlic are not completely known. In another study, it has been established that garlic components affect the N-acyltransferase activity of human bladder tumor cells (Chung, 1999).…”
