Effects of Estradiol on the Paracrine Regulator Expression of In Vitro Maturated Murine Ovarian Follicles

Abstract: The preservation of female germ cells is important in the individuals with ovarian dysfunction and failure. For this purpose, ovarian follicle in vitro maturation (OFIVM) is an important technology for the retrieval of mature oocytes. In the in vivo follicular development, paracrine factors such as angiotensin (AT) and anti-Müllerian hormone (AMH) play important roles. We attempted to add estrogen during the OFIVM and to assess their expression on the follicular cells. The ovaries and pre-antral follicles were… Show more

“…The methods used for in vitro culture of follicles have been taken from previous studies (Kim et al , , ,c). Non‐coated and alginate‐coated single and multiple follicles were placed in droplets (20–40 μl) of culture media on a culture dish (Falcon 353004, 60 × 15 mm; BD Biosciences, Irvine, CA, USA), one follicle or follicle cluster per droplet, 25 droplets per dish.…”

“…The ovary offers a safe storage space for oocytes within follicles (Y. Y. Kim et al, ; Y. Y. Kim, J. W. Yun et al, ; Ross, ; Tamadon, Park, Kim, Kang, & Ku, ). It releases steroid hormones after puberty and thereby fosters follicular development and periodic oocyte release (Y. J. Kim et al, ; Y. J. Kim, Ku, Rosenwaks et al, ; Y. J. Kim, K. E. Park et al, ). Ideally and anatomically, the artificial ovary needs to contain follicles isolated from cryopreserved ovarian tissue and possibly other ovarian cells to provide growth factors (Chiti et al, ).…”

“…Inflammatory responses can be evaluated through CD45 and F4/80 immunostaining (Laronda et al, ; Vanacker, Dolmans, Luyckx, Donnez, & Amorim, ). Ovarian functional recovery in mice can be validated with lowered serum follicle stimulating hormone concentration, elevated estrogen level, increased inhibin level, and pregnancy (Carroll & Gosden, ; Kim, Perez, et al, ; Kim, Park, et al, ; Y. Y. Kim, H. Kim, Ku et al, ; Laronda et al, ; Laronda et al, ; Smith et al, ).…”
