Abstract:Reactive oxygen species (ROS) are widely believed to cause or aggravate several human pathologies such as neurodegenerative diseases, cancer, stroke and many other ailments. Antioxidants are assumed to counteract the harmful effects of ROS and therefore prevent or treat oxidative stress-related diseases. In this report, recent human studies exploring the efficiency of antioxidants in prevention and treatment of various diseases are reviewed. Few antioxidants including edaravone (for ischemic stroke in Japan), Nacetylcysteine (for acetaminophen toxicity), alfa-lipoic acid (for diabetic neuropathy) and some flavonoids (polyphenolic compounds present in dietary plants), such as micronized purified flavonoid fraction (diosmin and hesperidin) and oxerutins (for chronic venous insufficiency) as well as baicalein and catechins (for osteoarthritis) have found accepted clinical use. However, despite much enthusiasm in the 1980s and 1990s, many well-known agents such as antioxidant vitamins and also more recently developed compounds such as nitrones have not successfully passed the scrutiny of clinical trials for prevention and treatment of various diseases. This has given rise to a pessimistic view of antioxidant therapy, however, the evidence from human epidemiological studies about the beneficial effects of dietary antioxidants and preclinical in vitro and animal data are compelling. We have probably wasted too much time on agents like antioxidant vitamins instead of focusing on more disease specific, target-directed, highly bioavailable antioxidants. We here discuss possible reasons for the lack of success in some clinical trials and seek to provide some suggestions to be considered if antioxidant therapy is to succeed as an effective therapeutic strategy.

Abstract:The focus in the research of antioxidants is notably displaced in favour of research of enzyme derivatives. Extracellular superoxide dismutase (EC-SOD) is of the particular interest, as it demonstrates in vivo the protective action against development of atherosclerosis, hypertension, heart failure, diabetes mellitus. The reliable association of coronary artery disease with the decreased level of heparin-released EC-SOD was established in clinical researches. To base and develop antioxidant therapy, various SOD isozymes, catalase (CAT), the methods of experimental gene therapy, combined application of SOD-and CAT-activities are used. Covalent bienzyme SOD -CHS -CAT conjugate (where CHS -chondroitin sulphate) showed in the experimental trials its high efficacy as the antithrombotic agent. Pre-clinical research data approve the reliable option to gain for it the status of drug candidate. The trend to use the components of glycocalyx and extracellular matrix for target delivery of medical substances is evident. Development of new enzyme antioxidants for therapeutical destination is closely connected with becoming and progress of medical biotechnology, pharmaceutical industry, bioeconomy.

Background: Edaravone (Eda) is a free-radical scavenger which is used in treating stroke, cerebral hemorrhage and some other diseases in clinic. However, it's antioxygenation during development is still unknown.Method: An oxidative damage model was established in chick embryo by using a generator of free radicals, 2,2'-azobis[2-methylpropionamidine] dihydrochloride (AAPH). The mortality rate and embryo's weight were measured to detect whether retarded growth happened. In order to observe the blood vessels, the yolk-sac blood vessels and CAM blood vessels density were observed. The malondialdehyde (MDA) content and superoxide dismutase (SOD) enzymatic activity were detected to evaluate the oxidative damage in the chick embryo.Result: In this model, embryo development and angiogenesis were heavily affected by AAPH. However, Eda alleviated the growth retardation and anti-angiogenesis induced by AAPH. After AAPH treatment, the MDA content increased and SOD enzymatic activity declined which was also mitigated by Eda. Conclusion:Overall, our research revealed that Eda mitigated embryonic anti-angiogenesis induced by oxidative damage after AAPH treatment, which may be helpful for future clinical studies.stress [7]. The chick embryo holds a wide range of advantages, with low cost and less ethical arguments [8]. It is a classical development model where the chick yolk sac and chorioallantoic membranes (CAMs) are extensively used in research related to angiogenesis. Besides, the growth cycle of chick embryo is short, where 21 days for chick embryo is similar to 10 months for human [9]. Moreover, it is a simple spinal biological, whose growth process can be easily observed [10].In order to mimic chronic oxidative stress exposed to human, 2,2'-azobis[2-methylpropionamidine] dihydrochloride (AAPH) was repeatedly treated in a small dose. AAPH is an azo compound and completely dissolved in alkyl, peroxyl and alkoxyl radicals at physiological temperature [11]. AAPH has been widely used to study the mechanism of oxidative stress [12,13]. In our previous experiments, the chick embryo on embryo development day (EDD) 9 was treated with a large dose of AAPH. After incubation for 24 hours, the chick embryo suffered from oxidative stress and the cardiovascular system was affected [7]. Nevertheless, whether chronic oxidative stress affects the development of embryos is still unclear. In our study, AAPH was injected to chick embryos in long-term and the development of chick embryo was observed.
