Effects of different cytokinins on the shoot regeneration from apple leaves of 'Royal Gala' and 'M.26'

Abstract: The effects of different types of cytokinins on the shoot regeneration from leaf explants of apple scion 'Royal Gala' and apple rootstock 'M.26' were evaluated. Regeneration media contained either thidiazuron, or 6-benzylaminopurine, or meta-topolin, or zeatin, or kinetin, or their N9-ribosides, respectively, in the concentration range 0.5 to 8.0 mg 1-1. Effects of 'these cytokinins were evaluated on the percentage of regeneration (R%) and that of vitrification (V%) and on the number of regenerated shoots per … Show more

“…However, the predominant type of cytokinin depended on genotype (Kamboj et al 1999). Furthermore, changes induced by cytokinins were also described both at the tissue and cell level (Dobránszki et al 2004). Pre-treatment of apple shoots in liquid medium with TDZ prior to regeneration also had a wide range of effects on leaf cells (Sriskandarajah and Goodwin 1998), and the structural characteristics of leaves altered by cytokinins are related to the organogenic ability of explants .…”

“…The classical cytokinins, namely N 9 -ribosides (applied at lower concentrations of 17-23 lM than nonribosides at 22-30 lM), showed the same or better regeneration, except for BAR. The OI was highest when TOPR was applied at 21.44 lM for 'M26' (Dobránszki et al 2004). The developmental pattern of shoot regeneration of 'M26' under different cytokinins is presented in Fig.…”
