Plant tissue culture has become one of the fundamental tools of plant science research. It is extensively employed in the production, conservation and improvement of plant resources. The presence of somaclonal variation in populations derived from tissue culture is affecting the use of tissue culture negatively and has remained a major problem. Conversely, it is a source of new desirable clones/variants with better agronomic traits. In this review, we summarize the possible causes, detection methods and desirability of variants. Somaclonal variation is one of the most researched and reviewed topics. Hence, we restricted ourselves to outlining various examples which may be used as important references for researchers who intend to identify and/or characterize somaclonal variants while using tissue culture for research and production. Emphasis is placed on the negative effects of somaclonal variation. However, this review also includes examples of some useful variants generated as a result of somaclonal variation.
BackgroundMedicinal plants are possible sources for future novel antioxidant compounds in food and pharmaceutical formulations. Recent attention on medicinal plants emanates from their long historical utilisation in folk medicine as well as their prophylactic properties. However, there is a dearth of scientific data on the efficacy and stability of the bioactive chemical constituents in medicinal plants after prolonged storage. This is a frequent problem in African Traditional Medicine.MethodsThe phytochemical, antioxidant and acetylcholinesterase-inhibitory properties of 21 medicinal plants were evaluated after long-term storage of 12 or 16 years using standard in vitro methods in comparison to freshly harvested materials.ResultsThe total phenolic content of Artemisia afra, Clausena anisata, Cussonia spicata, Leonotis intermedia and Spirostachys africana were significantly higher in stored compared to fresh materials. The flavonoid content were also significantly higher in stored A. afra, C. anisata, C. spicata, L. intermedia, Olea europea and Tetradenia riparia materials. With the exception of Ekebergia capensis and L. intermedia, there were no significant differences between the antioxidant activities of stored and fresh plant materials as measured in the β-carotene-linoleic acid model system. Similarly, the EC50 values based on the 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay were generally lower for stored than fresh material. Percentage inhibition of acetylcholinesterase was generally similar for both stored and fresh plant material. Stored plant material of Tetradenia riparia and Trichilia dregeana exhibited significantly higher AChE inhibition than the fresh material.ConclusionsThe current study presents evidence that medicinal plants can retain their biological activity after prolonged storage under dark conditions at room temperature. The high antioxidant activities of stable bioactive compounds in these medicinal plants offer interesting prospects for the identification of novel principles for application in food and pharmaceutical formulations.
The study compared mineral, chemical and antioxidant qualities of Cleome gynandra, a wild leafy vegetable, with two widely consumed commercial vegetables, Brassica oleracea and Beta vulgaris. Mineral nutrients were quantified with inductively coupled plasma mass spectrometry (ICP-MS), phenolic compounds using ultra-high performance liquid chromatography coupled to a mass spectrometer (UHPLC-MS) and β-carotene and vitamin C using high performance liquid chromatography with a photodiode array detector (HPLC-PDA). The antioxidant potential was evaluated using 2,2–diphenyl−1–picryl hydrazyl (DPPH) and oxygen radical absorbance capacity (ORAC) assays. Cleome gynandra had highest concentrations of phosphorus, potassium, calcium, iron, zinc, ascorbic acid, total phenolics, and flavonoids; whereas sodium, magnesium, manganese, copper and β-carotene were higher for B. vulgaris. The significantly higher antioxidant activity (P ≤ 0.05) exhibited by C. gynandra in comparison to the two commercial vegetables may be due to its significantly high levels of vitamin C and phenolic acids. These findings on the mineral, chemical and antioxidant properties of C. gynandra provide compelling scientific evidence of its potential in adding diversity to the diet and contributing toward the daily nutritional requirements of millions of people for food and nutritional security.
Since the discovery of topolins as naturally occurring aromatic cytokinins (CKs), they have emerged as genuine alternatives to the long serving CKs such as benzyladenine, zeatin and kinetin in plant tissue culture (PTC). Globally, the past 15 years has witnessed a surge in the use of topolins and their derivatives in research laboratories. Topolins, especially the meta-topolin and its derivatives have been employed for culture initiation, protocol optimization and for counteracting various in vitro induced physiological disorders in many species. Evidence from various studies indicate the rising popularity and advantages (although not universal for all species) of topolins compared to other CKs. In this review, we assess the use of topolins in PTC with emphasis on their metabolism, structure-activity relations and effect on morphogenesis in vitro. In addition, the review provides a detailed list of species that have been used to study the effect of topolins in comparison with other CKs, the growth parameters affected and recommended concentrations are also provided.
Issues relating to food availability, accessibility/affordability, and food utilization remain paramount among different stakeholders such as policymakers and academics. Using data from 250 maize farming households in Nigeria, the study used Foster–Greer–Thorbecke and probit regression model to investigate the factors determining households food security. The food insecurity measure shows that 23.2% points of the households express the incidence of food insecurity while 5.5% points and 1.8% points were found to have depth and severity of food insecurity, respectively. After controlling for households’ socio-economic and demographic characteristics, the probit regression model suggested that, among others, value of output sold, education, credit access and participation in government safety nets program significantly influenced food security among the maize farmers in the study area. Based on our findings, effort should be intensified to enhance the productivity of land through improved production practices. There should be high-level awareness that will increase farmers’ participation in safety net programs. Thus, government at all levels (local, state, and federal) should have adequate budget allocation to this course in order to improve the livelihood outcomes of the farming households.
