Effects of Dietary ω-3 and ω-6 Lipids and Vitamin E on Serum Cytokines, Lipid Mediators and Anti-DNA Antibodies in a Mouse Model for Rheumatoid Arthritis

Abstract: It is clear from our observations that the beneficial effects of FO can be enhanced by the addition of 500 IU of vit-E in the diet. The FO diet containing 500 IU of vit-E may specifically modulate the levels of IL-6, IL-10, IL-12 and TNF-alpha and thereby may delay the onset of autoimmunity in the MRL/lpr mouse model. The observations from this study may form a basis for selective nutrition intervention based on specific fatty acids and antioxidants in delaying the progress of RA.

“…Some investigators have suggested that a low lipid diet may be useful to attenuate the exocrine gland inflammation in this autoimmune disorder (Di Giacinto & Piergiacomi, 1991). In contrast, other researchers have proposed that dietary supplementation with omega-3 and other polyunsaturated Nutrient intake in Sjögren's syndrome JM Cermak et al fatty acids may help to reduce the titers of anti-DNA antibody titers, decrease the generation of pro-inflammatory cytokines, delay the onset and lessen the severity of Sjögren's syndrome, SLE and=or RA (Fernandes et al, 1990;Chandrasekar & Fernandes, 1994;Shapiro et al, 1996;Calder, 1997;James & Cleland, 1997;Alexander, 1998;Reifen et al, 1998;Venkatraman & Chu, 1999). It is possible that some of the dietary differences observed in this study may have been due to variations between groups in height, size and body mass index.…”

“…Based on preclinical data, it was postulated that the combination of these nutrients would produce synergistic or additive effects without compromising safety (Chilton et al, 1993;Venkatraman & Chu, 1999). An odorless liquid nutritional supplement was developed as a vehicle to enhance compliance.…”

“…22 Similar to our study in which IL-4 production was elevated, it has been reported that feeding (n-3) PUFA can lower IL-4 levels, in a rodent model of contact dermatitis local. 39 Although the literature is inconsistent, there is some evidence that suggests dietary FO can lower serum IL-10 levels in a model of experimental rheumatoid arthritis, a state with high serum IL-10 40 and can lower IL-10 in response to allergen in susceptible neonates. 41 In this study, feeding FO did not significantly modify IL-2 production in MLN cells stimulated with a polyclonal T-cell mitogen (ConA), indicating that increasing dietary (n-3) PUFA does not suppress T-cell function in obese JCR:LA-cp rats, which has been reported in healthy animals.…”
