In this study, the mitochondrial damage effect and mechanism of zearalenone (ZEA) in swine small intestine IPEC-J2 cells in vitro were comprehensively characterized. The analyses revealed that ZEA at high doses (8 and 7 μg/mL) can significantly increase P < 0.05 the malondialdehyde levels and decrease antioxidant enzymes activities after 48 h of exposure. Meanwhile, the reactive oxygen species (ROS) accumulation increased in high dose ZEA-treated groups after 2 h treatment, but decreased due to the ROS-induced mitochondrial damage and the caused cell apoptosis after 48 h of high does ZEA treatment. Moreover, the decreasing of mitochondrial membrane potential (MMP; ΔΨ) in high dose ZEA exposure was observed in line with the increasing ROS production in mitochondria. Results suggest that ZEA exposure can induce mitochondrial damage by reducing antioxidant enzyme activities, accumulation of ROS, and decreasing MMP. The mitochondrial damage had a dramatic concentration-effects relationship with ZEA.

This study demonstrates that low doses (somewhat above the No Observed Adverse Effect Level, NOAEL) of the mycoestrogen zearalenone (ZEN) and its metabolites display multispecificity towards various biological targets in gilts. The observed responses in gilts were surprising. The presence of ZEN and zearalenols (ZELs) did not evoke a response in the porcine gastrointestinal tract, which was attributed to dietary tolerance. Lymphocyte proliferation was intensified in jejunal mesenteric lymph nodes, and lymphocyte counts increased in the jejunal epithelium with time of exposure. In the distal digestive tract, fecal bacterial counts decreased, the activity of fecal bacterial enzymes and lactic acid bacteria increased, and cecal water was characterized by higher genotoxicity. The accompanying hyperestrogenism led to changes in mRNA activity of selected enzymes (cytochrome P450, hydroxysteroid dehydrogenases, nitric oxide synthases) and receptors (estrogen and progesterone receptors), and it stimulated post-translational modifications which play an important role in non-genomic mechanisms of signal transmission. Hyperestrogenism influences the regulation of the host’s steroid hormones (estron, estradiol and progesteron), it affects the virulence of bacterial genes encoding bacterial hydroxysteroid dehydrogenases (HSDs), and it participates in detoxification processes by slowing down intestinal activity, provoking energy deficits and promoting antiporter activity at the level of enterocytes. In most cases, hyperestrogenism fulfils all of the above roles. The results of this study indicate that low doses of ZEN alleviate inflammatory processes in the digestive system, in particular in the proximal and distal intestinal tract, and increase body weight gains in gilts.

Środki spożywcze i materiały paszowe pochodzenia roślinnego są bardzo często wektorami niskich dawek mikotoksyn fuzaryjnych (28). Ze względu na szeroki zakres ich zastosowań w przemyśle spożywczym i w produkcji pasz, a także wysoki udział produktów zbożowych w diecie człowieka i zwierząt są ważnym elementem odpowiedzialnym za stan zdrowia ludzi oraz wydajność procesu produkcyjnego zwierząt (10). Najczęściej spotykanymi mikotoksynami fuzaryjnymi są zearalenon (ZEN) oraz deoksyniwalenol (DON).Zearalenon i jego metabolity powodują zaburzenia w rozrodzie zwierząt, ponieważ w swej aktywności są podobne do estradiolu (40). α-ZEL jest dominującym metabolitem ZEN występującym w organizmie świń, podczas gdy u innych gatunków zwierząt, jak np. u brojlerów, krów i owiec dominującym produktem metabolizmu ZEN jest β-ZEL o dużo mniejszej aktywności metabolicznej (9). Efekt aktywności ZEN zależy od nasilenia i charakteru procesów metabolicznych mających miejsce w organizmie zwierząt (18). Zależy również od statusu immunologicznego (5, 20) układu rozrodczego, który podlega ciągłym zmianom na skutek zmian hormonalnych wynikających z okresu dojrzewania płciowego, etapu cyklu rozrodczego czy ciąży. Wykazano, że ZEN może być immunotoksyczny w niskich stężeniach (33). W odpowiedzi na obecność mikotoksyny, wrażliwe komórki przechodzą wiele zmian w zakresie istotnych procesów metabolicznych (proliferacji i różnicowania się komórek, apoptozy, syntezy cząsteczek biologicznie aktywnych, itp.) (37). Pomimo że badania nad ZEN zostały ostatnio zintensyfikowane, to istnieje niewiele badań nad jego wpływem na barierę jelitową i potencjalnego związku pomiędzy ekspozycją na ZEN a stanami zapalnymi jelit (13).
