Effects of daptomycin in combination with other antimicrobial agents: a review of in vitro and animal model studies

Abstract: This review summarizes the in vitro and animal model data available on antibiotic combinations with daptomycin. The majority of studies focus on the clinically relevant combinations of daptomycin with rifampicin or with gentamicin. These studies demonstrate that daptomycin does not adversely affect the activity of other antimicrobial agents that may be administered concomitantly. Overall, additive or indifferent effects with daptomycin combinations were observed; however, synergy was observed for certain isola… Show more

“…Whether even higher, off-label daptomycin doses would increase efficacy in the treatment of VRE bacteremia, without increasing toxicity, also remains to be explored. Similarly, the effect of proposed strategies of combination treatment with daptomycin and ampicillin (31) or rifampin (34) could not be assessed adequately from these data.…”

“…Several in vitro and animal infection models have shown additive or synergistic effects with DAP combination therapy (29,42,44). Previous work has examined the effect of combination exposures on the emergence of DAP resistance, although these studies are often limited by a number of factors, including short exposure duration, use of static antibiotic concentrations, testing of reference strains rather than clinical isolates, and the small number of combinations examined (4,20,35,38).…”

“…Eight antimicrobials that have been used therapeutically in combination with DAP were selected (29,42,44). In addition to measuring changes in the DAP MIC, strains selected under these antimicrobial combinations were examined for phenotypic changes associated with DAP resistance, including changes to cell wall thickness (CWT) and cell membrane physiology.…”
