Effects of clonidine on breathing during sleep and susceptibility to central apnoea

Abstract: We hypothesized that administration of clonidine would decrease the hypocapnic apnoeic threshold (HAT) and widen the CO2 reserve during non-REM sleep. Methods Ten healthy subjects (4 females) (age 22.3±3.0 years; BMI 25.5±3.4 kg/m2) were randomized to receive placebo or 0.1 mg/45Kg of clonidine on 2 separate nights. Ventilation and upper airway resistance were monitored during wakefulness and sleep. Two separate experiments were performed: Protocol 1 (n=8), CO2 reserve, HAT and HcVR were determined using non-… Show more

“…21 Clonidine enhances both sensory and motor block from epidural local anaesthetic. 22 At clinical doses it has minimal effects on hypercapnic and hypoxic ventilatory drive in adults [23][24][25][26] and children. 27 Cognitive impairment, difficulties with verbal communication, and the application of below-knee plasters did not allow accurate assessment of dermatomal spread of the epidural block.…”

“…A single report describes a potential role for clonidine as adjunctive therapy for hypocapnic central sleep apnea syndromes [182]. In a study of 10 healthy subjects (4 females) (age 22.3 ± 3.0 years; BMI 25.5 ± 3.4 kg/m 2 ), subjects were randomized to receive placebo or 0.1 mg/45 kg of clonidine on 2 separate nights.…”
