Background: Caffeic acid phenethyl ester (CAPE) is a phytochemically active component obtained from honeybee hive propolis. CAPE has been reported to show antimitogenic, anticancer, and other beneficial medicinal properties. Many of its activities have been reported to be mediated by inhibiting levels of matrix metalloproteinase, that is, MMP-2 and MMP-9. We hypothesize the effect of CAPE on the metastasis of colon cancer cells in both in vitro and in vivo. Methods: Cell migration, motility, invasion were evaluated also expression of protein and matrix metalloproteinases (MMPs) such as MMP-2 and MMP-9 were measured in SW-480 cancer cells in vitro. The cells were exposed to Phorbol 12-myristate 13-acetate (PMA) and were treated with various concentration of CAPE. Results: The treatment of CAPE caused significant decrease (P<0.05) in both cell motility and invasion. The treatment of CAPE inhibited activity of MMP-2 and MMP-9 and their protein with increasing dose in SW-480 cancerous cells. Antimetastatic activity was evaluated in vivo in BALB/c mice by injecting them with CT-26 mouse colon cancer cells via tail vein and were treated with CAPE (20 mg/kg) orally for 21 days. The CAPE treatment significantly (P<0.05) reduced count of pulmonary nodules. The mice showed decreased plasma MMP-2 and MMP-9 activity after 21 days treatment with CAPE. Conclusion:The study suggested beneficial role of CAPE in preventing invasion of colon cancer and metastasis via MMP-2 and MMP-9 mediated pathway.

AbstractÖz Purpose: The esophageal burns caused by corrosive substances are of great importance since they progress along with serious complications. Here, we have aimed to evaluate our 39 cases we followed up and treated in our clinic due to taking in corrosive substances according to their clinical characteristics and our treatment approaches. Materials and Methods: Thirty nine patients whose file records could be obtained and who were treated for corrosive esophagitis between January 2000 and December 2007 were included in our study. Clinical features and treatment of those cases were recorded and evaluated. Results: The intake rate of corrosive substances was higher in women. The amount of corrosive substance taken in was prominently higher in those having it for suicidal purposes compared to those having it by accident. According to their endoscopic features, 82.1% of the cases were found to have had endoscopic findings referred to as normal or grade 1. While ulcer grade 2-3 comprised 10.3% of all the cases, grade 4 comprised 7.7% of all the cases with severe burns. Conclusion: To stop the oral intake, performing an endoscopic evaluation and making a treatment plan in accordance with the endoscopy results for the patients applying the clinic in the early stage due to the intake of corrosive substances are of great importance in terms of preventing the complications likely to occur. Amaç:Koroziv maddelerin oluşturdukları özofagus yanıkları ciddi komplikasyonlar ile seyrettiği için çok önemlidir. Bu çalışmada kliniğimizde takip ve tedavi ettiğimiz koroziv madde içen 39 olgumuzu klinik özellikleri ve tedavi yaklaşımlarımıza göre değerlendirmeyi amaçladık. Gereç ve Yöntem: Çalışmamıza Ocak 2000-Aralık 2007yılları arasında Selçuk Üniversitesi, Meram Tıp Fakültesi,Göğüs Cerrahi kliniğinde tedavi edilen ve dosya kayıtlarına ulaşılabilen 39 hasta dahil edildi. Bu hastaların klinik özellikleri ve tedavileri kaydedilerek değerlendirildi. Bulgular: Koroziv madde içme oranı kadınlarda daha yüksek idi. İçilen koroziv madde miktarı suisit amaçlı içenlerde yanlışlıkla içenlere oranla belirgin yüksek idi. Endoskopik özelliklerine göre olguların %82.1'inde normal veya evre 1 endoskopi bulgusuna rastlanılmıştır. Evre 2-3 ülser tüm olguların %10.3'ünü oluştururken, evre 4 ağır yanık tüm olguların %7.7'ini oluşturuyordu. Sonuç: Koroziv madde kullanımı sonrası erken dönemde başvuran hastalarda oral alımın kesilmesi, erken evrede endoskopik değerlendirmenin yapılması ve endoskopi sonucuna göre tedavi planının yapılması komplikasyonların önlenmesi açısından büyük öneme sahiptir.

burn is a common health problem in the pediatric age group and causes serious esophageal injuries. The medical treatment in acute phase of corrosive esophageal injury is of particular importance for prevention of esophageal stricture. We therefore aimed to investigate the possible beneficial effect of trapidil (triazolopyrimidine), an inhibitor for phosphodiesterase and platelet-derived-growth-factor, during acute phase of esophageal corrosive injury. Wistar albino rats were randomly allocated to untreated, treated, and sham-operated groups (n = 10 for each group). Corrosive esophageal burn was generated with 10% NaOH solution. The rats were left untreated (untreated group) or treated with trapidil as a single dose of 40 mg/kg intraperitoneally after one hour of the injury (treated group). Abdominal esophageal segment was isolated and tied in sham-control group. The studied esophageal segment was removed from each animal after 24 hours. Malondialdehyde (MDA) and nitric oxide (NO) levels were measured in the esophageal tissues. The ulcer depth was graded by histopathologic examination. MDA and NO levels were significantly higher in the untreated group than in the treated group. Namely, trapidil treatment significantly decreased MDA and NO levels in the injured tissues, the levels of which are similar to those in the tissues of control animals. The grades of ulcer depth were significantly improved in the treated group. These results indicate that the reactive oxygen radicals increase in the early phase of corrosive esophagitis and cause tissue damage.
