Effects of Benzylidenecyclopentanone Analogues of Curcumin on Histamine Release from Mast Cells

Abstract: Curcumin [1,7-bis(4-hydroxy-3-methoxyphenyl)-1,6-heptadiene-3,5-dione] is a natural yellow pigment isolated from the rhizomes of the plant Curcuma longa L. (turmeric). Its chemical structure is described in Fig. 1.1) The pharmacological effects of curcumin include anti-inflammatory, 2) antioxidant, 3) antiprotozoal, 4) antibacterial, 5) anticancer, 6) antihepatotoxic, 7) hypolipidemic, 8) and antifibrotic effects. 9) Curcumin showed no toxicity in acute toxicity studies in rats, guinea pigs, and monkeys, 10) a… Show more

“…Compound 48/80 also stimulates histamine release from RPMCs in both the presence and absence of extracellular calcium. The present study demonstrates that curcumin potently suppressed histamine release probably through the inhibition of the degranulation process following a rise in intracellular Ca 2+ levels, in accordance with previous reports (Matsuda et al, 2004; Suzuki et al, 2005; Nugroho et al, 2009). …”

“…When CUR was ingested after exercise, the increase in CK was significantly lower and CK reduced faster than PLA after 5 days of exercise (Figure B). It has been reported that CUR has a membrane protective effect and inhibits production of histamine and prostaglandin (PG), which are involved in vascular hyperpermeability . In addition, the structure of the membrane changes depending on the blood CUR concentration .…”

“…19) Curcumin were also found to decrease histamine release from mast cells. 20) Additionally, IL-10 gene therapy could ameliorate the acute phase of EAM by stabilizing mast cells. This therapeutic mechanism is exerted by the binding of IL-10 to specific IL-10 receptors located on mast cells, leading to prevention of the release of inflammatory mediators.…”
