Effect of vitamin K2 administration on depression status in patients with polycystic ovary syndrome: a randomized clinical trial

Abstract: Background Patients with Polycystic ovary syndrome (PCOS) are predisposed to the development of several mental comorbidities such as depression. According to several studies, PCOS can be managed by improving insulin sensitivity. The insulin-sensitizing effect of vitamin K has been reported in recent studies. Therefore, in the current trial, we assessed the effect of administrating vitamin K2 (Menaquinone-7) on depression status in women afflicted with PCOS. Metho… Show more

“…48,[111][112][113][114] Vitamin K2, which improved depression in a randomized controlled trial of patients with depression and polycystic ovarian syndrome, is also hypothesized to mechanistically achieve effects via modification of insulin resistance. 115 While one study of pioglitazone suggested its antidepressant effects were only present in individuals with insulin resistance, it is unknown whether putative antidepressant effects of SGLT2 inhibitors may be dependent on diabetes diagnosis. 113 SGLT2 inhibitors share some characteristics with other antidiabetic medications, but are singular in their capacity to reliably induce ketogenesis.…”

“…PPAR-gamma agonist pioglitazone and metformin have been studied in bipolar and unipolar depression, with some (but not all) studies reporting positive results 48,111–114 . “…A trial of 84 women with PCOS allocated to vitamin K2 supplements, which is proposed to have insulin-sensitizing effects, or placebo found significant reductions in BDI-II scores in the intervention arm (mean 15.0 AE 8.3 vs. 16.9 AE 7.9, p ¼ 0.013). 62 However, vitamin K's glycemic effects in the general population are controversial. 63 A randomized placebo-controlled trial of the effect of 12 weeks of 100 mg/day of coenzyme Q10 (CoQ10) in 55 women with PCOS found significant reductions in BDI scores (p ¼ 0.03) and high-sensitivity C-reactive protein (p ¼ 0.005) as well as biochemical androgen levels compared to placebo.…”
