Effect of Thiocyanate on Human Skin Collagenase Activity

Abstract: Using tissue culture, human skin collagenase activity and its relationship to serum collagenase inhibitors and the effect of thiocyanate was studied. The following was found: 1) Collagenase activity did not become apparent in the culture medium before 48 hours; it reached a peak on the 4th day, gradually decreased, and then became elevated again on the 7th day of culture. 2) Using the immunodiffusion technique, α2‐macroglobulin was detected up to the 4th day and α1‐antitrypsin until the 7th day. 3) On the 2nd… Show more

“…This preparation was freeze-dried after extensive dialysis against cold water. Preparation of a1I(I) and a2 of pig skin collagen These collagens were purified from pig skin by the method of Shinkai et al (1974). In brief, 30ml of acid-soluble collagen (2.3 mg/ml in 0.06M-sodium acetate buffer, pH4.8) was centrifuged at lOOOOOg for 1 h and the resulting supernatant was denatured at 50°C for 20min.…”

“…Furthermore, thinning of the dermis in rat skin during catagen has been reported from several sources (see Griesemer 1956). Shinkai et al (1974) and Reynolds et al (1977) have shown the dissociation of collagenase inhibitorcomplex by SCNin vitro. Reddick et al (1974) have demonstrated collagenase in dermal fibroblasts and in what the authors suggested to be an extracellular location in the dermis of human skin; the extracellular location of collagenase has also been reported by Gross (1976).…”

“…Hollander et al (1949) reported the loss of scalp hair in a human being who was treated with SCN-for hypertension. Shinkai et al (1974) and Reynolds et al (1977) suggested the activation of latent collagenases by SCN-in vitro, andHanstein (1969, 1970) discussed the chaotropic properties of the ion.…”

“…Preparation andfractionation ofcollagen Tumor tissue was separated from epidermis and subcutaneous tissue and homogenized with Ultra Turrax Ganke & Kunkel KG, Staufen, West Germany) in 0.5 M sodium acetate then stirred for 48 hours at 4°C to extract serum components. Neutral and acid soluble collagens were prepared and purified as previously described (7). Pepsin treated collagens were isolated from the residue after extraction with 0.5 M acetic acid according to the method of Chung & Miller (8).…”
