Effect of the Citrus Flavone Nobiletin on Circadian Rhythms and Metabolic Syndrome

Abstract: The importance of the circadian clock in maintaining human health is now widely acknowledged. Dysregulated and dampened clocks may be a common cause of age-related diseases and metabolic syndrome Thus, circadian clocks should be considered as therapeutic targets to mitigate disease symptoms. This review highlights a number of dietary compounds that positively affect the maintenance of the circadian clock. Notably the polymethoxyflavone nobiletin has shown some encouraging results in pre-clinical experiments. A… Show more

“…It can be concluded from the few studies presented that intaking a high-energy source at nighttime and its diet component may contribute to impaired energy and glucose metabolism and reproductive hormone disruption during pregnancy ( 126 , 127 ). In pregnant animals, it was found that disturbed intake timing, or feeding (during the day period by nocturnal rodents), gave rise to an irregular circadian rhythm in the mother and fetus ( 127 ) and was capable of altering the microbiota profiles to give rise to metabolic disorders ( 128 ). However, studies that translate these mechanisms to pregnant women have not yet been carried out.…”

“…Some of the potentials as chronotherapeutic agents are flavonoids. Nobiletin, a polymethoxy flavone obtained from dried citrus peel, known as Citri Reticulatae Pericarpium fruit, was identified as a particularly effective clock amplitude-enhancing small molecule and can directly affect the mammalian circadian system ( 128 130 ). Time-restricted feeding partially restores diurnal rhythms of the ileal microbiome ( 131 ).…”

“…Several in vitro and in vivo studies have shown that flavonoids, especially flavanones, the main class of flavonoids in citrus extracts, such as naringin and naringenin [ 22 , 28 ], possess antiproliferative, anti-inflammatory, and pro-apoptotic properties [ 29 , 30 , 31 , 32 , 33 , 34 ]. Other compounds found in citrus fruits that have demonstrated potential anticancer effects include quercetin [ 15 , 35 , 36 , 37 , 38 , 39 , 40 , 41 ], hesperidin [ 42 , 43 , 44 ], hesperetin [ 45 , 46 ], polymethoxyflavones [ 47 , 48 , 49 ], eriodyctiol [ 50 , 51 ], bergapten [ 52 , 53 ], tangeretin [ 54 , 55 ], auraptene [ 56 , 57 , 58 ], limonin [ 59 ], naringenin [ 60 , 61 , 62 , 63 , 64 , 65 ], and naringin [ 66 , 67 , 68 ], as shown in ( Table 1 ). These findings suggest a potential role for flavonoids in cancer therapy, including breast cancer.…”

“…Nobiletin, an important component of polymethoxylated flavones isolated from citrus fruit peels, has been extensively investigated in various diseases, such as diabetes, cancer, nonalcoholic fatty liver disease, and sepsis (7). Recently, nobiletin has received increased attention because of its superior physical functions, which exhibit minimal toxicity and a variety of pharmacological benefits, including anti-inflammatory, anti-oxidant stress, anti-cancer, and anti-diabetic effects (8,9).…”
