Effect of Streptozotocin-induced Diabetes on Pancreatic Insulin Content of the Fetus

Abstract: Diabetes was produced in five-day pregnant Sprague- Dawley rats by intravenous injection of 40 mg./kg. body weight of streptozotocin. Normal pregnant rats were used as controls. Pancreases of the mothers and their offsprings were extracted by acid-alcohol and the insulin measured by immunoassay using rat insulin as a standard. Nonfasting blood sugar of the streptozotocin-treated pregnant rats was 285 ± 18 mg./100 ml. and their pancreatic insulin concentration was 5.9 ± 1 μg./gm. compared to 103 … Show more

“…Pancreatic insulin content has been found to be unchanged [90,103], increased [89,104], or decreased [105][106][107], The same discrepancy was observed regarding plasma insulin concentration which was reported to be enhanced, lowered, or unchanged [re viewed in 65]. These conflicting data must be related, for a large part, to the difficulty of controlling hyperglycemia induced by diabe togenic drugs.…”

“…Work on the rat and rabbit has shown that offspring of experimental diabetic mothers have hyperfunction of the endocrine pancreas (Gepts, 1957;Kim, Runge, Wells & Lazarow, 1960; Rishi, Golob, Becker & Shah, 1969;Wellman, Wölk, Lazarus & Branceto, 1969;Golob, 1970;Pitkin & Van Orden, 1974). The aims of our study were to investigate the morphological and functional behaviour of the rat foetal and neonatal endocrine pancreas in experimental diabetes and to correlate these findings with our data in man.…”

“…6 In the streptozocin-induced diabetic pregnant rat, the placenta is consistently larger and the fetus smaller than in controls. 7 - 10 The placenta of the rat is complex, having many distinct regions; even in the labyrinthine portion of the chorioallantoic placenta, the trophoblast is three layered. Previous biochemical studies 7 have demonstrated continued cellular division and increased glycogen content in these diabetic placentas, consistent with a functional immaturity of this organ.…”
