Effect of Single Injection of Pentylenetetrazole in a Subconvulsive Dose on Cl-Conductance of the GABAA-Receptor Complex

Abstract: Muscimol-stimulated Cl(-)conductance in brain cortex synaptoneurosomes from Wistar rats decreased 15 min after single intraperitoneal injection of pentylenetetrazole in a subconvulsive dose. These changes reflected a decrease in functional activity of the GABA(A)-receptor/Cl(-)ionophore complex. Muscimol-stimulated Cl(-)conductance in synaptoneurosomes returned to normal 48 h after pentylenetetrazole administration.

“…Changes in functional activity of GABA A -RC in synaptoneurosomes of rat brain cortex at different stages of PTZkindling were demonstrated (including our previous results, Fig. 2) [5][6][7][8]. The basal and muscimolstimulated 36 Cl -entry into synaptoneurosomes increased at the early stage of kindling.…”

“…These data can be explained if we compare them with our previous results. Single injection of PTZ in the subconvulsive dose inhibited muscimol-stimulated Cl -conduction of synaptoneurosomes isolated from the cerebral cortex of animals 15 min after PTZ injection, which attests to reduced functional activity of GABA A -RC; muscimol-stimulated Cl -conduction of synaptoneurosomes is completely restored 48 h after PTZ injection [6]. During the early stage of kindling, when convulsions have not yet develop, basal and muscimol-stimulated 36 Cl -entry into synaptoneurosomes increased [7].…”

“…We previously detected changes in the GABA A -RC Cl -conduction after a single dose of the convulsant (pentylenetetrazole; PTZ) in the subconvulsive dose and at the early stage of kindling development (before appearance of convulsions) [6,7].…”

“…It is reported that pentylenetetrazol has an important role in development of epileptic seizures via mechanism of blocking GABAA receptors (Rebrov et al, 2004). The extracts of S. marianum seeds are rich in many phytochemicals especially phenols and flavonoids (Atraqchi et al, 2014), which are thought to exert anti-convulsion effects by modulating channels (Diniz et al, 2015).…”
