Effect of puerarin on melanogenesis in human melanocytes and vitiligo mouse models and the underlying mechanism

Abstract: Puerarin is the major bioactive ingredient derived from the root of the Pueraria lobata (Willd.), and its antioxidative stress effects have been demonstrated in several previous studies. Moreover, Puerarin can upregulate melanin synthesis and microphthalmia-associated transcription factor (MITF) transcription by increasing cAMP level of intracellular cyclic adenosine monophosphate. Vitiligo is an acquired cutaneous disorder of pigmentation, and the pathogenesis has remained elusive.Current treatment modalities… Show more

“…In the 4-benzyloxyphenol-induced vitiligo mouse model, after one week of Puerarin Treatment, HE staining of the skin at the depigmented sites showed increased hair follicles, which were surrounded by a large number of melanocytes. Puerarin at 40 μmol/L significantly increased the melanin content of human melanocytes by decreasing the phosphorylation of ERK in the cells to promote TRP-1 and MITF expression, which led to an increase in melanin content ( Ding et al, 2019 ).…”

“…The melanin granules formed in the basal layer are transferred and stored in the overlying keratinocytes [1,2] . Dysfunctional melanogenesis or the absence of melanocytes can lead to skin hypopigmentation [3] .…”
