Most pharmacokinetic studies on anthelmintic drugs have been performed on non-parasitized animals. However, it seems likely that the parasite burden could influence the deposition of such drugs. The pharmacokinetics of moxidectin administered orally and by subcutaneous injection was compared in lambs exposed to nematode infection and in parasite naive lambs. Plasma samples were analyzed for moxidectin over 40 days post-treatment. The main pharmacokinetic parameters calculated demonstrated a significant change in drug deposition in infected lambs when compared to controls. The area under the plasma concentration-time curve was decreased 54% and 46% by infection in the subcutaneous and oral groups, respectively. There was also a major decrease in the mean residence time in parasitized lambs. In parallel, the clearance of the drug was increased by infection. Thus, parasite infection dramatically influences the disposition of moxidectin in lambs. These results may contribute to determining a therapeutic strategy adapted to heavily infested animals.

ABSTRAKPemberian antihelminth setengah dosis merupakan metode sederhana untuk deteksi dini terjadinya resistensi parasit yang menginfeksi ruminansia kecil. Ketika kegagalan antihelminth secara tunggal pada domba asli dari Indonesia, kombinasi antihelminth dari kelas kimia dengan mekanisme yang berbeda sebagai salah satu strategi alternatif pengendalian parasit. Penelitian ini membandingkan efektivitas ivermectin (IVM) dan albendazole (ABZ) baik yang diberikan secara terpisah dengan pemberian satu dan setengah dosis maupun dikombinasikan pada domba secara yang alami terinfeksi Ostertagia. , and control groups. The treatment effectiveness was determined using the Faecal egg count reduction (FECR) on before and after 7, 14, 21, 28, 35, and 42 days of treatment. FECR of half-dose IVM, full-dose IVM, half-dose ABZ, full-dose ABZ, and both combined ranged from 78100, 96100, -7189, -52164, and 54100, respectively. The results showed that full-dose IVM was the most effective to against Ostertagia until 42 days and followed the combined that effective until 28 days. Half-dose and full-dose ABZ weren't effective to against Ostertagia.

This article reviews the literature dealing with the effects of composition and quality of diet and feeding time on the pharmacokinetics and efficacy of some anthelmintic drugs in ruminants. Studies have suggested that greater availability, and therefore improved anthelmintic activity, is possible through temporary feed restriction. It is also recommended that anthelmintic drugs should not be given to animals whilst they are maintained on large feed intakes, particularly of lush pasture that promotes rapid gastric transit, as this may reduce drug availability and anthelmintic efficacy. Generally, feeding animals low-quality fibrous diets reduces the passage rate of digesta and allows more time for absorption of several anthelmintic drugs and their metabolites from the gut. Some kinetic data of drugs given to animals on such diets may be slightly different, but this does not necessarily indicate alteration of the dosages of the anthelmintic drug. Nonetheless, due consideration should be given to anthelmintic dosages under various dietary regimes if optimum efficacy is to be achieved at all times.
